摘  要:目的探讨不同浓度的去整合素Kistrin对兔眼后发性白内障(Posterior capsular opacification,PCO)发生过程中晶状体上皮细胞(lensepithelial cells,LECs)增殖的抑制作用,以明确Kistrin的眼内有效浓度。方法制备兔眼PCO模型,行白内障摘除术,术毕3个实验组前房分别注入40μg/L、80μg/L、160μg/L的Kistrin,对照组前房注入林格氏液。术后在裂隙灯显微镜下观察PCO发生情况及形态,于术后14 d及3个月时取材,分别检测晶状体囊膜上LECs增殖细胞核抗原(proliferating cell nuclear antigen,PCNA)的表达。结果术后第14天实验组LECs的PCNA表达均较对照组降低(P<0.05);第3个月时仅80μg/L组较对照组明显降低(P<0.05);两个时间点80μg/L组LECs的PCNA表达均较其余实验组明显降低(P<0.05),40μg/L组与160μg/L组间无显著性差异(P>0.05)。同一浓度术后第14天与3个月时LECs的PCNA表达比较,仅80μg/L组差异无显著性,余三组比较差异有显著性。结论 80μg/L Kistrin术后早期及3个月均可有效地抑制兔眼PCO的发生,可作为进一步研究去整合素抑制PCO形成研究的较适宜浓度。Objective To explore the inhibition of disintegrin Kistrin in the proliferation of lens epithelial cells when rabbits' eyes occurred posterior capsule opacification(PCO) for 14 d and 3 months, so as to provide experimental and theoretical basis for Kistfin in anti- PCO mechanisms. Methods The anterior chamber of the rabbits PCO models in experimental group were injected different concentrations (40,80,160μg/L) of Kistrin at the end of the operations and those in control group were injected ringer's solution. The occurrence and morphology of PCO were observed by slit-lamp microscope. Then the eyeballs were removed at 14 d and 3 months to detect the expression of proliferating cell nuclear antigen (PCNA)in LECs. Results After 14 d, LECs PCNA expression in experimental groups was lower than that in control group ( P 〈 0. 05 ). After 3 m, it was lower only in 80 μg/L group ( P 〈 0. 05 ). At the two points in time, PCNA expression of 80μg/L Kistrin group was all lower than that of rest experimental groups ( P 〈 0. 05 ), between which was not significandy different( P 〉 0.05 ). And only in 80 μg/L group, PCNA expression at the two points had no difference( P 〉 0.05 ). Conclusions 80 μg/L Kistrin can effectively suppression the PCO formation in early stage and 3 months after operation. 80 μg/L is a candidate concentration for Kistrin in studying its inhibition of PCO formation.
