埃兹蛋白与喉癌侵袭与转移相关性的研究

摘　　要：目的探讨埃兹蛋白在喉癌中的表达以证实其与喉癌侵袭与转移的相关性。方法采用逆转录聚合酶链反应(reverse transcription-polymerase chain reaction,RT-PCR)技术和免疫组织化学技术(immunohistochemistry)检测埃兹蛋白在58例喉癌组织、癌周非癌组织和25例喉癌转移淋巴结中的表达。结果 RT-PCR显示埃兹蛋白在喉癌组织中的表达(0.506±0.245)较癌周非癌组织中的表达(0.079±0.082)明显增高,二者之间的差异具有统计学意义(P<0.01)。在25例转移淋巴结中的表达(0.651±0.299)高于喉癌组织,二者之间的差异具有统计学意义(P<0.05)。免疫组化显示埃兹蛋白在癌周非癌组织、喉癌和颈部转移淋巴结中的表达依次增高(P<0.05),其表达强度与喉癌的临床TNM分期、T分级、颈淋巴结转移和病理学分级有关(P<0.05),而与患者的性别、年龄以及吸烟程度无关(P>0.05)。结论埃兹蛋白的高表达与高侵袭能力具有相关性,可能在喉癌的侵袭和转移过程中发挥重要作用。Objective To study the expression of Ezrin in laryngeal carcinoma for confirming its correlation with invasion and metastasis of laryngeal carcinoma.Methods The expression of Ezrin in 58 no-cancer mucosa of pars laryngea,laryngeal carcinoma and 25 metastatic lymph nodes were evaluated by reverse transcription polymerase chain reaction（RT-PCR） and immunohistochemistry.Results The result of RT-PCR showed that the expression of Ezrin（0.506±0.245）in laryngeal carcinomas was obvious higher than that in the normal mucosa around cancer（0.079±0.082）（P〈0.01）;In 25 metastatic lymph nodes,the expression of Ezrin（0.651±0.299） was higher than that in carcinoma（P〈0.05）.The results of immunohistochemistry showed that the expression of Ezrin increased in no-cancer mucosa of pars laryngea,laryngeal carcinoma and metastatic lymph nodes by turns（P〈0.05）,which was related with TNM stage,T grading,the metastasis of cervical glands and pathology grading of laryngealcancer（P〈0.05）,but no relation with gender,age and smoking of patients（P〈0.05）.Conclusion Higher expression of Ezrin is related to the invasion ability of laryngeal carcinoma,which maybe play an important role in the process of invasion and metastasis of laryngeal carcinoma.
