摘  要:目的探讨血小板衍生生长因子-A(PDGF-A)及其受体(PDGFR-α)3、羧基磷脂酰肌醇激酶(PI3K)和磷酸化蛋白激酶B(p-AKT)在成釉细胞瘤(AB)中的表达及意义。方法应用免疫组化法检测9例牙胚、36例成釉细胞瘤中上述4种蛋白的表达。结果 PDGF-A表达在牙胚与滤泡型AB、滤泡型AB与丛状型AB之间比较有显著差异(P<0.05);PDGFR-α表达在棘皮瘤型AB、颗粒细胞型AB和无细胞变异型AB之间比较有显著差异(P<0.05);PI3K表达在牙胚组与丛状型AB和无细胞变异型AB比较、滤泡型AB与丛状型AB比较差异均有显著性(P<0.05);p-AKT表达在牙胚组分别与AB、丛状型AB和无细胞变异型AB比较均有显著差异(P<0.05)。结论牙源性组织中PDGF-A/PDGFR-α功能的发挥可能通过PI3K/AKT信号传导通路的介导实现。PDGF及其受体的高表达及PI3K/AKT信号传导通路异常活化与成釉细胞瘤的发生、发展有关。Objective To investigate the expression and its significance of platelet-derived growth factor-A(PDGF-A) and its receptor(PDGFR-α),PI3K and p-AKT in ameloblastomas(ABs).Methods The expressions of above-mentioned 4 proteins in 9 tooth germs and 36 Abs were detected by using immunohistochemistry(IHC).Results PDGF-A expression was significantly different between tooth germs and plexiform ABs,plexiform ABs and follicular ABs(P〈0.05);PDGFR-α expression was significantly different among acanthomatous subtype,granular subtype and non-cellular variation ABs(P〈0.05);PI3K expression was significantly different among tooth germs,plexiform ABs and non-cellular variation ABs,and between plexiform and follicular ABs(P〈0.05);p-AKT expression was significantly different between tooth germs and ABs,plexiform ABs and non-cellular variation ABs respectively(P〈0.05).Conclusion Function exertion of PDGF-A/PDGFR-α in odontogenic tissues could be completed which was mediated by PI3K/AKT signal transduc-tion pathway.Genesis and development of AB is correlated with high expression of PDGF and its receptor,and abnomal activation of PI3K/AKT signal transduction pathway.
