摘  要:[目的]探讨肿瘤坏死因子TNF-β+252基因多态性与肺癌遗传易感性的关系。[方法]采用聚合酶链式反应-限制性片段长度多态性(PCR-RELP)方法对152例肺癌患者(病例组)和127例健康者(对照组)的TNF-β+252基因型进行分析。[结果]TNF-β+252基因型频率和等位基因分布频率在肺癌组和对照组比较差异均无统计学意义(χ2=0.60,P=0.74;χ2=0.24,P=0.63)。分层分析发现,不吸烟人群中肺癌组和对照组基因型分布差异有统计学意义(χ2=6.67,P=0.04),与AA基因型相比GG基因型增加了肺癌的患病风险(OR:3.97,95%CI:1.34~11.79),而等位基因的分布频率在两组之间差异无统计学意义(χ2=1.54,P=0.22)。[结论]TNF-β+252多态性与肺癌易感性相关,不吸烟人群中GG基因型增加了肺癌的发病风险。[Objective]To investigate the association between polymorphisms in human tumor necrosis factor-β(252)gene and susceptibility of lung cancer.[Methods]Polymerase chain reaction-restrained fragment length polymorphism technique(PCR-RFLP)was applied to detect the single nucleotide polymorphisms(SNPs)of TNF-β+252 in 152 lung cancer patients(case group)and 127 healthy people(control group).[Results]No significant differences were found in the distribution of TNF-β+252 genotypes and allele frequencies between lung cancer patients and controls(χ2 = 0.60,P = 0.74;χ2 = 0.24,P = 0.63).After stratification analysis according to smoking,it revealed that the TNF-β+252 GG genotype in no-smokers subgroups were associated with significantly increased risk of lung cancer comparing the AA genotype(χ2 = 6.67,P = 0.04).But there was no significant difference in the distribution of A and G two alleles frequencies of TNF-β+252 GG between the case group and control group(χ2 = 1.54,P = 0.22).[Conclusion]TNF-β+252 gene polymorphism may be associated with the susceptibility of lung cancer and GG genotype may increase the risk of lung cancer among non-smoking people.
