摘  要:目的四种含RGD的细胞粘附肽(RGD、RGD(NH2)2、RGDS、RGDS-NH2)对人胃癌细胞BGC823(Human stomach cancer BGC823)生长增殖的影响,进而探讨含四种RGD诱导BGC823细胞凋亡的作用。方法通过MTT法检测四种含RGD对BGC823细胞生长增殖的抑制作用;通过HE染色、透射电子显微镜、免疫组织化学、流式细胞术和凋亡细胞的DNA片段分析等方法观察凋亡细胞的形态结构变化及细胞凋亡的机制。结果含RGD能抑制BGC823细胞的生长增殖,并呈浓度依赖性;其含RGD诱导BGC823细胞凋亡可能通过线粒体引发这一途径。结论含RGD诱导BGC823细胞凋亡,从而抑制肿瘤细胞的增殖。并且在介导细胞凋亡中,RGD具有高度保守的特性。Objective To investigate the effect of the RGD-containing cellular adhesion tripeptides on human stomach cancer BGC823,and to study the function of the RGD tripeptide in inducing BGC823 apoptosis.Methods BGC823 cells were treated with the RGD tripeptides at various concentrations.MTT assay was used to examine the growth and proliferation of BGC823 cells after treatment with the RGD tripeptides for 48 hours.HE staining and electromicroscope were used to observe the morphology of apoptotic cells,and Survivin,flow cytometry were also used to analyze the BGC823 apoptosis.Results the RGD tripeptides significantly inhibits the growth and proliferation of BGC823 cells in dose-dependent manner.Conclusion the RGD-containing cellular adhesion tripeptide may inhibit tumour cell growth and proliferation through inducing apoptosis.Induce apoptosis caused by mitochondria.In these processes,RGD is of high conservation.
