针刺对血管性痴呆大鼠学习记忆能力及海马内乙酰胆碱酯酶含量的影响

摘　　要：目的复制拟血管性痴呆(vascular dementia,VD)模型,探讨针刺百会、人中、大椎穴对大鼠学习记忆能力及血清和海马内乙酰胆碱酯酶(acetylcholinesterase,AchE)含量的影响。方法用双侧颈总动脉缺血再灌注法复制VD大鼠模型,将VD大鼠随机分为模型组、针刺组和西药组。采用大鼠跳台试验检测学习记忆能力;采用酶联免疫法检测血清和海马组织AchE含量。结果针刺组大鼠跳台反应期时间比模型组显著缩短(P<0.05);针刺组大鼠血清和海马AchE含量显著低于模型组(P<0.05)。结论降低VD大鼠血清和海马内AchE含量是针刺的作用机制之一。Objective To establish vascular dementia（VD） model in an attempt to study the effects of acupuncture at Baihui,Renzhong,and Dazhui acupoints on learning and memory abilities and on the levels of acetylcholinesterase（AchE） in the serum and the hippocampus of VD rats.Methods The VD model was induced by ischemia and reperfusion of bilateral common carotid artery.The VD rats were randomized into model group,acupuncture treated group and western medicine treated group.The abilities of learning and memory were detected by jumping stair test,and the levels of AchE in the serum and the hippocampuswere assayed using enzyme-linked immunosorbent assay.Results The response time of jumping stair test was significantly shortened in the acupuncture treated group compared with that in the model group（P〈0.05）;the levels of AchE in the hippocampus and the serum in the acupuncture treated group were significantly decreased compared with those in the model group（P〈0.05）.Conclusion The decrement of AchE levels in the serum and the hippocampus is one of underlying mechanisms for acupuncture in the treatment of VD rats.
