摘  要:目的观察姜黄素诱导人急性早幼粒细胞白血病HL-60细胞凋亡的作用,并探讨作用机制。方法用台盼蓝计数观察姜黄素对HL-60细胞生长的抑制作用,AnnexinV-FITC/PI染色流式细胞仪检测细胞凋亡,RT-PCR检测凋亡相关基因mRNA的表达,Western blot检测细胞内NF-κB p65蛋白的表达。结果姜黄素对HL-60细胞生长有抑制作用,并诱导细胞凋亡。姜黄素作用前后HL-60细胞中抗凋亡基因Bcl-2和Bcl-xl表达无明显变化;促凋亡基因Bax和Bid表达增高;P53基因均不表达;NF-κB mRNA和蛋白表达均增高,并呈剂量依赖性。结论姜黄素能诱导HL-60细胞凋亡和上调促凋亡基因Bax和Bid,这可能与NF-κB表达上调有关。Objective To explore the effect and mechanism of curcumin on apoptosis of myeloblastic leukemia HL-60 cells.Methods After HL-60 cells treated with curcumin,the cell growth inhibition was detected by trypan blue count,the apoptosis was detected by flow cytometry through the staining of AnnexinV-FITC/PI,the expression of genes about apoptosis were detected by RT-PCR,the expression of NF-κB p65 protein was analyzed by Western blot.Results Curcumin could inhibit the proliferation and induce the apoptosis of HL-60 cells.The mRNA expression of Bax,Bid and NF-κB in HL-60 cells treated with curcumin were higher than those without curcumin,while no difference between Bcl-2 and Bcl-xl,and no expression of P53 in both HL-60 cells treated with and without curcumin.Curcumin could up-regulate the expression of NF-κB p65 protein in HL-60 cells with a concentration-dependent manner.Conclusion Curcumin could induce HL-60 cells apoptosis and increase the expression of Bax and Bid,possible mechanism was up-regulation of NF-κB.
