摘 要:目的:测定阿魏酸、川芎嗪的体外回收率,确定其校正方法,并进行体内稳定性考察。方法:利用高效液相色谱仪测定微透析样品中阿魏酸、川芎嗪的浓度,并计算探针回收率;把探针植入大鼠颈静脉,考察探针在10 h内和5只大鼠之间体内回收率的稳定性。结果:1.在相同流速下,相对回收率与药物浓度无关,并且在相同条件下相对回收率与相对损失率近似相等。2.5只大鼠之间探针的回收率之间有差异性;3.探针在大鼠体内10 h内回收率能保持相对稳定。结论:当每次实验都进行反透析法回收率校正时,所建立的微透析取样技术可用于川芎嗪、阿魏酸的动物在体药动学研究。AIM:To investigate the recovery in vitro and stability in vivo of microdialysis probes with tetrameth-yl-pyrazine/ferulic acid as model drugs.METHODS: The concentration of tetramethylpyrazine and ferulic acid was detected by HPLC.Stability of the recovery was studied under the condition of probe planted into five rats’ jugular veins at the time interval of 10 h.RESULTS: 1.The concentration had no obvious effect on relative recove-ry,there was no significant difference between relative recovery and relative loss in the same flow rate.2.The RL were different in different conditions.3.The relative loss had good stability in 10 hours in vivo.CONCLUSION:Under the relative recovery of every experiment is determined by retrodialysis,miecodialysis sampling can be used for the pharmacokinetic study of tetramethylpyrazine/ferulic acid.
