脑缺血再灌注大鼠外周血EPCs与VEGF、bFGF、eNOS水平变化及相关性

摘　　要：目的探讨大鼠脑缺血再灌注后外周血内皮祖细胞(endothelial progenitor cells,EPCs)、血清血管内皮生长因子(vascular endothelial growth factor,VEGF)、碱性成纤维细胞生长因子(basic fibroblast growth fac-tor,bFGF)及内皮型一氧化氮合成酶(endothelial nitric oxide synthase,eNOS)水平变化及相关性。方法 SD大鼠随机分为正常空白对照组、假手术组、脑梗死组、糖尿病组、糖尿病假手术组和糖尿病缺血再灌注组。链脲佐菌素诱导制作糖尿病模型,线栓法制作脑缺血再灌注模型。流式细胞仪测定EPCs数量,ELISA法测定VEGF、bFGF及eNOS的数量。结果脑梗组EPCs、VEGF、bFGF和eNOS数量较正常组及假手术组增高(P<0.01),糖尿病脑梗组EPCs较糖尿病组下降(P<0.05),而VEGF、bFGF和eNOS数量则升高(P<0.05),EPCs与VEGF和bFGF数量变化有相关性(P<0.05),与eNOS数量变化相关性无统计学意义。结论大鼠脑缺血再灌注后外周血EPCs与VEGF和bFGF水平变化存在相关性,它们可能共同影响着缺血性脑血管病的一些病理生理过程。这种变化及相关性对于估测脑梗死患者病情程度和预后有一定的指导意义。【Objective】 To investigate the changed levels of peripheral blood endothelial progenitor cells（EPCs） and serum VEGF,bFGF,eNOS in rats with cerebral ischemic reperfusion injury and their correlation.【Methods】 SD rats were randomly divided into the normal group,the normal sham-operation control group,the cerebral ischemic reperfusion injury group,the diabetic group,the diabetic sham-operation control group and the diabetic cerebral ischemic group.The diabetic models were induced by streptozotocin（STZ）.The cerebral ischemic rats were operated with the thread occlusion method.The number of peripheral blood EPCs was detected by flow cytometry.The levels of serum VEGF、bFGF、eNOS were detected by enzyme-linked immunosorbent assay（ELISA）.【Results】 The number of peripheral blood EPCs and the levels of serum VEGF,bFGF,eNOS were higher in the cerebral ischemic reperfusion injury group than the normal group and normal sham-operation control group（P 0.01）,The number of peripheral blood EPCs was lower in the diabetic cerebral ischemic group than the diabetic group（P 0.05）,but the levels of serum VEGF,bFGF,eNOS in the diabetic cerebral ischemic group were higher than the diabetic group（P 0.05）.The number of peripheral blood EPCs and the levels of serum VEGF、bFGF have a certain correlation（P 0.05）.But the correlation of the number of peripheral blood EPCs and the levels of serum eNOS have not a statistically significance.【Conclusion】 There is a correlation between the number of peripheral blood EPCs and the levels of serum VEGF,bFGF in rats with cerebral ischemic reperfusion injury,they may affect the pathological and physiological process of the ischemic cerebrovascular disease together,this correlation may has guide meaning for estimating the state of an illness of cerebral infarction.
