摘 要:目的研究乳腺浸润性导管癌、乳腺良性病变组织中垂体瘤转化基因(PTTG),基质金属蛋白酶-14(MMP-14)和血管内皮生长因子-C(VEGF-C)的表达水平,探讨其临床病理意义。方法 40例乳腺浸润性导管癌、20例乳腺良性病变手术切除标本常规石蜡包埋切片,PTTG、MMP-14和VEGF-C染色方法为免疫组化SP法。结果乳腺浸润性导管癌PTTG、VEGF-C和MMP-14表达阳性率(75.0%、85.0%和77.5%)明显高于乳腺良性病变(20.0%、30.0%和25.0%,P<0.01),有淋巴结转移病例PTTG、VEGF-C和MMP-14表达阳性率明显高于无淋巴结转移病例(P<0.01),而在不同年龄、肿瘤大小、分化程度组间差别无统计学意义(P>0.05)。乳腺浸润性导管癌中PTTG与VEGF-C和MMP-14表达呈高度一致性(r=0.586,0.657,P<0.01),VEGF-C与MMP-14表达亦呈高度一致性(r=0.649,P<0.01)。结论 PTTG、VEGF-C和MMP-14的表达可能是反映乳腺浸润性导管癌发生、发展、生物学行为及预后的重要生物学指标,三者有望成为乳腺癌抗淋巴管治疗的新靶点。【Objective】 To investigate the expressions of pituitary tumor transforming gene(PTTG),matrix metalloproteinase-14(MMP-14) and vascular endothelial growth factor-C(VEGF-C) in human breast invasive ductal carcinoma and benign lesions.Try to explore its clinicopathologic significance.【Methods】 Immunohistochemical method was used to detect the expressions of PTTG,MMP-14 and VEGF-C in the routinely paraffinembeded sections of breast invasive ductal carcinoma(n =40) and benign cases(n =20).【Results】 The positive rates of PTTG,VEGF-C and MMP-14 were significantly higher in breast invasive ductal carcinoma(75%,85%,77.5%) than in benign lesions(20%,30%,25%,P 0.01).There were statistical relationships between their expressions and TNM stage and lymph node metastasis(P 0.01).But there were no significant differences between the expressions of PTTG,VEGF-C,MMP-14 and age,size and differentiation of tumor(P 0.05),The expression of PTTG was positively correlated with VEGF-C and MMP-14 expressions.【Conclusion】 The expression of PTTG,VEGF-C and MMP-14 might be important biological markers reflecting the carcinogenesis,progression,biological behaviors and prognosis of breast invasive ductal carcinoma;they can be used as new targets of antilympha-tic therapy for breast invasive ductal carcinoma.
