摘 要:目的检测DD3 mRNA在前列腺癌中的表达,探讨其在前列腺癌诊断中的作用,并分析其与前列腺癌临床分期和病理分级之间的关系。方法采用SYBR-Green I实时荧光定量RT-PCR法检测DD3mRNA在34例前列腺癌、20例前列腺增生组织及3种前列腺癌细胞株中的表达,同时收集临床病例数据,分析其表达含量与临床分期和病理分级之间的关系,应用SPSS13.0对数据进行统计分析。结果①DD3 mRNA在前列腺癌组织和前列腺癌细胞株中的表达量明显升高,与其在前列腺增生组织中的表达量相比,差异具有显著性(P<0.001)。②在不同的临床分期与病理分级中,DD3 mRNA的表达量随临床分期和病理分级增加而增加,其差异亦具有显著性(P<0.001)。结论 DD3 mRNA在前列腺癌中呈高表达,可作为前列腺癌的一项特异性早期诊断标记物,其与前列腺癌的临床分期和病理分级具有相关性,提示其有可能在前列腺癌的发生发展中起重要作用。【Objective】 To observe the expression of DD3 mRNA in prostate cancer and analyze its correlation with clinical stage and pathological grade of prostate cancer.【Methods】 SYBR-Green I quantitative real-time RT-PCR was used to detect the expression of DD3 mRNA in 34 prostate cancer tissues,20 benign prostatic hyperplasia tissues and 3 kinds of prostate cancer cell lines.The relationship between DD3 and the clinical-pathological parameters in prostate cancer were evaluated.Data were analyzed with SPSS 13.0 statistical software.【Results】 Expression of DD3 mRNA in prostate cancer tissues and prostate cancer cell lines was significantly higher than in benign prostatic hyperplasia tissues(P 0.001).With the increasing of clinical stage and pathologic grade in prostate cancer,DD3 mRNA expression levels were significantly increased simultaneously(P 0.001).【Conclusion】 The results of this study indicate potential utility of DD3 mRNA as a early diagnostic biomarker for prostate cancer.Its correlation with clinical stage and pathological grade suggests it may play an important role in the development and progression in prostate cancer.
