摘　　要：目的改进Migrastatin母核的全合成工艺。方法以(4R,5R)-二甲基-2,3-邻异丙亚基-L-酒石酸酯为起始原料,经DIBALH还原、Pb(OAc)4催化重排、LiBH4还原、Dies-Alder反应、Ferrier重排以及水解生成关键中间体,再经GrubbsⅡ催化分子内二烯烃关环反应生成Migrastatin母核。结果与讨论经11步反应,Migrastatin的总收率为1.8%。OBJECTIVE To improve the total synthetic processing of Migrastatin.METHODS The target compound was synthesized from L-tartaric acid.After diisobutylaluminium hydride reduction,Pb（OAc）4 under the action of the rearrangement,LiBH4 reduction,4＋2 Dies-Alder reaction,the Ferrier rearrangement under camphorsulfonic acid,hydrolysis,the key intermediate was generated.After the cyclization reaction of the key intermediate catalysed by Grubbs Ⅱ,the Migrastatin was produced.RESULTS and CONCLUSION Migrastatin was synthesized by an eleven-step reaction in total yield of 1.8%.
