摘　　要：目的观察microRNA-133a(miR-133a)在乳腺癌、乳腺良性病变及正常乳腺组织中的表达,探讨miR-133a的表达与乳腺癌临床病理特征的关系及其对乳腺癌发生发展的意义。方法采用组织芯片平台,应用原位杂交方法检测90例乳腺癌组织、26例乳腺良性病变组织和34例癌旁正常乳腺组织miR-133a表达。结果乳腺癌miR-133a阳性率(26.7%)低于乳腺良性病变(53.8%)和癌旁正常组织(44.1%)(P<0.05);乳腺癌组织miR-133a的表达与腋淋巴结转移(P<0.05)及TNM高分期(P<0.05)有关联,与患者年龄、肿瘤大小、组织学分级、雌激素受体(ER)、孕激素受体(PR)和HER-2/neu人表皮生长因子受体2(c-erbB-2)过表达均无相关性(P>0.05)。结论 miR-133a在乳腺癌组织中表达下调,提示其可能作为重要的抑癌因子参与乳腺癌的发生和发展过程,并可能是乳腺癌转移的潜在标志。Objective To investigate the expression of miR-133a in breast carcinoma and its clinicopathologic significance.Methods miR-133a expression was detected in 90 cases of breast carcinoma and 26 cases of benign breast lesions and 34 cases pericancerous normal breast tissues which were at least 4 cm away from the carcinoma and contained no histological abnormalities.Results The expression rate of miR-133a in breast carcinoma was significantly lower than that of the benign lesions and pericancerous breast tissue（P0.05）.There were correlations of miR-133a expression between lymph node metastasis（P0.05） and TNM stage（P0.05）,whereas no correlations were found among the expression of miR-133a and patient age,tumor size,histological grading,ER,PR and c-erbB-2 status（P0.05）.Conclusion The decreased expression of miR-133a in breast carcinoma implies its role of a tumor suppressor factor in the pathogenesis and progression.It might be a potential marker for breast carcinoma metastasis.
