OPN和NF-κB/p65在乳腺癌中的表达及临床意义

摘　　要：目的观察OPN、NF-κB/p65在乳腺癌组织中的表达情况,并分析其与病理学分级、淋巴结转移和激素受体表达等的关系以及两者的相关性。方法选取病理证实的80例乳腺癌手术切除组织标本,40例良性乳腺组织标本,用免疫组织化学法(EliVision法)检测OPN、NF-κB/p65的表达水平。结果 OPN和NF-κB/p65在80例乳腺癌组织中的高表达率分别为63.8%(51/80)和61.3%(49/80),在40例良性乳腺组织中的高表达率分别为15.0%(6/40)和20.0%(8/40),OPN、NF-κB/p65分别在乳腺癌组织与良性乳腺组织中高表达率的差异均有统计学意义(P<0.05);OPN和NF-κB/p65的高表达率与其病理分级、腋窝淋巴结转移有关(P<0.05),其中NF-κB/p65的高表达率亦与HER-2表达有关(P<0.05);OPN和NF-κB/p65与组织学类型、TNM分期、肿瘤大小、ER和PR无关(P>0.05);乳腺癌组织中OPN与NF-κB/p65的表达呈正相关(P<0.05)。结论乳腺癌组织中OPN和NF-κB/p65的高表达率较高,与乳腺癌的侵袭性生物行为密切相关,两者联合检测可能成为乳腺癌转移及预后的重要指标。Objective To evaluate the expression of osteopontin（OPN）and NF-κB/p65 in breast cancer,and its relationship with histological grading,lymph node metastasis,the expression of hormonal-receptor as well as the correlation of them.Methods Immunohistochemical（Eli Vision）method was used to detect the expression of OPN and NF-κB/p65 in eighty pathological confirmed and surgical resected breast cancer,and forty noncancerous breast tissue.Results The high positive rates of the expression of OPN and NF-κB/p65 in breast cancer were 63.8%（51/80）and 61.3%（49/80）.The high expression of them in noncancerous breast tissue were 15.0%（6/40）and 20.0%（8/40）.The high expression level of OPN and NF-κB/p65 had correlation with histological grading,axillary node metastasis（P0.05）,and that of NF-κB/p65 also had correlation with the expression of HER-2（P0.05）,but had no correlation with histological grading,stage of TNM,size of breast cancer,ER,PR and HER-2（P0.05）.OPN was positively correlated with NF-κB/p65 in breast cancer（P0.05）.Conclusion The high expression of OPN and NF-κB/p65 in breast cancer is higher than that in noncancerous breast tissue,which is correlated with the biological behavior of breast cancer,and a joint detection of them might be an important marker and prognostic factor in breast cancer.
