摘 要:在改性Fe3O4纳米颗粒存在的条件下用乳液聚合法制备了表面带有环氧基的聚甲基丙烯酸缩水甘油酯-聚甲基丙烯酸甲酯P(GMA-co-MMA)磁性复合微球,对其粒径、形态、磁含量、磁性质进行了表征;在复合微球表面引入氨基后,以戊二醛为交联剂,固定木瓜蛋白酶,探讨了给酶量、戊二醛浓度对固定化酶活性的影响,考察了酶学性质。结果表明,该复合微球的平均粒径为170nm,具有超顺磁性,磁含量可以控制在20%~50%之间;当戊二醛浓度为5%,给酶量为200mg时,达到固定化酶最适使用条件,其在热、酸碱和操作稳定性上均优于游离酶。Poly(glycidyl methacrylate)-co-poly(methyl methacrylate) P(GMA-co-MMA) magnetic composite microspheres with epoxy groups on their surfaces were prepared by emulsion polymerization in the presence of oleic acid-coated Fe3O4 nanoparticles as magnetic resources.The morphologies,diameter,magnetic content and magnetic properties of the resulting composite microspheres were identified.Amino groups were introduced onto surfaces of the magnetic microspheres through surface modification.Papain was immobilized on magnetic microspheres by cross-linked reaction with glutaraldehyde.And the effects of amount of enzyme and concentration of glutaraldehyde on the activity of immobilized enzyme were studied.The properties of immobilized enzyme were also investigated under different temperature and pH.The results show superparamagnetic microspheres have a mean size of 170 nm and the magnetic content can be controlled from 20% to 50%.The optimum immobilized conditions reached when the amount of enzyme was 200mg and concentration of glutaraldehyde was 4%.Compared with free papain,the immobilized papain possessed good pH and thermal stability,the durability has been improved as well.
