摘 要:目的以阿魏酸为主要指标,建立HPLC含量测定方法,为三棱药材的质量控制提供依据。方法三棱药材分别经甲醇提取和甲醇-2%氢氧化钠溶液(95∶5)提取水解后,采用HPLC法测定游离阿魏酸和总阿魏酸。色谱柱为Dia-monsil-C18(250 mm×4.6 mm,5μm);流动相为甲醇-1%冰醋酸(68∶32)洗脱;流速1.0 mL/min;柱温30℃;检测波长320 nm。结果阿魏酸在3.952×10-3~3.952×10-2μg线性关系良好,平均回收率分别为99.12%,RSD=1.74%和99.02%,RSD=4.54%。结论所建方法简单可靠,为三棱药材的质量控制提供了科学依据。OBJECTIVE To establish an HPLC method for the determination of ferulic acid in Sparganium stoloniferurm.METHODS HPLC was used to assay free ferulic acid and total ferulic acid in Sparganium stoloniferum which was extracted by methanol and methanol-2% NaOH in water(95∶5) solvents respectively.A Diamonsil-C18(250 mm×4.6 mm,5 μm) was used with a mixture of methanol-1% acetic acid(68:32) as the mobile phase.The flow rate was 1.0 mL/min and the column temperature maintained at 30 ℃.The detective wave length was 320 nm.RESULTS The linearity range of ferulic acid was 3.952×10-3~3.952×10-2μg.The average recovery of ferulic acid was 99.12%,the RSD of 1.74% and 99.02 % with the RSD of 4.54%.CONCLUSION This method can be used as the quality control standard for Spaganium stoloniferurm.
