摘 要:目的探讨三氧化二砷(As_2O_3)对人肝癌SMMC-7721细胞生长及其侵袭转移能力的影响。方法 MTT法检测不同浓度As_2O_3对人肝癌SMMC-7721细胞生长的影响,之后检测经As_2O_3作用前后对基底膜黏附能力的影响;Transwell膜侵袭系统观察As_2O_3作用前后SMMC-7721细胞游走与穿透基底膜能力的改变。结果 As_2O_3能显著抑制人肝癌SMMC-7721细胞的生长以及对Matrigel的黏附能力,相同浓度不同作用时间比较及相同作用时间不同浓度比较,差异均有统计学意义(P<0.01);As_2O_3能抑制细胞游走与穿透基底膜的作用,与对照组比较,游走穿膜细胞数及侵袭穿膜细胞数均减少(P<0.05)。结论 As_2O_3具有抑制人肝癌细胞生长增殖以及侵袭转移的能力。Objective To investigate the effect of arsenic trioxide(As2O3) in growth and adhesion as well as invasion metastasis of SMMC-7721 in human hepatocarcinoma in vitro.Methods Applying the MTT method to study the effect of growth and adhesion by As2O3 on SMMC-7721 cell.The transwell chamber membrane invasion culture system was used to study the invasion and metastasis.Results The As2O3 could significantly inhibit the SMMC-7721 cell growth,and inhibit the adhesion function with Matrigel.There was statistical difference between the similar action time with different concentration and the similar concentration with different action time(P〈0.01 ).The As2O3 could inhibit the cell migration through the basement membrane and movement function.Compared with the control group,the number of migration cell and invasion through the basement membrane was decreased(P〈0.05).Conclusion The As2O3 had the ability to inhibit the hepatocarcinoma cell growth as well as adhesion and invasion metastasis.
