摘  要:目的探讨去甲斑蝥素(NCTD)对人真皮淋巴管内皮细胞(HDLEC)增殖、迁徙的抑制以及诱导凋亡的作用,以寻找一种安全、有效、实用的“抗淋巴管生成”制剂。方法通过HDLEC二维培养,应用MTT法计算NCTD对HDLEC的抑制率:应用Hoechst33258荧光染色、划线法和倒置显微镜、透射电镜检测、观察NCTD对HDLEC增殖、迁徙的抑制和诱导凋亡作用。结果MTT法测得不同浓度和时间的NCTD对HDLEC的增殖有明显抑制作用(P〈O.01),其半数有效浓度(IC50)为0.0068mg/ml;应用Hoechst染色、倒置显微镜、透射电镜以及划线法,可观察到NCTD对HDLEC的形态和细胞的增殖具有显著的抑制和直接杀伤作用,并能促进、诱导HDLEC凋亡,其作用随NCTD浓度而加强(P〈0.01),同时可见NCTD对HDLEC的迁徙亦有明显的抑制作用(P〈0.01)。结论NCTD对HDLEC的增殖和迁徙有明显的抑制作用,且有剂量依赖性。Objective To investigate the effects of norcantharidin (NCTD) on proliferation and migration of human dermal lymphatic endothelial cells (HDLEC) in vitro. Methods HDLECs were treated with different concentration of NCTD in two-dimensional culture. The half inhibitory effective concentration (IC50) and cell toxic effect of NCTD on HDLECs were evaluated by MTT method. The inhibitory effects of NCTD on proliferation and migration of HDLEC, and the direct killing and apoptosis-inducing effects were measured and observed by Hoechst staining, scraping line method, inverted microscope and transmission electron microscope, respectively. Results MTT method showed that NCTD inhibited the proliferation and growth of HDLEC significantly and had direct-killing effect of HDLEC(P〈0.01), with IC50 of 0.006 8 mg/ml. Hoechst staining and scraping line demonstrated maeked inhibitory effects of NCTD on proliferation and migration of HDLECs (P〈0.01), the direct-killing and apoptosis-inducing effects of NCTD on HDLEC(P〈0.01), and the morphologic changes of HDLEC apoptosis, with dose-dependent. Conclusion NCTD has marked inhibitory effects on proliferation and migration of HDLEC and direct-killing and apoptosis-inducing effects in a dose-dependent manner.
