摘  要:[目的]观察大蒜素亚慢性染毒对大鼠脾体系数的影响以及停止染毒后的恢复状况,并对脏/脑系数的毒理学意义进行初步探讨。[方法]分别以3.3、10.0、30.0 mg/kg(以大蒜素计)灌胃方式给予动物,每日1次,连续90 d。分离脾脏、肝、肾、脑等组织称重。剩余动物停止灌胃后,继续观测45 d,处理方式同前。[结果]连续染毒90 d后,雄性实验组动物脾脏重量增大,呈现剂量-反应关系,中、高剂量动物脾脏重量、脾体系数与对照组比较差异有统计学意义(P<0.05或<0.01)。动物停止染毒45 d后,雄性中、高剂量组动物脾脏重量、脾体系数仍较对照组高(P<0.05)。在观察期内,各实验组动物脑重基本一致,与对照组比较差异无统计学意义(P>0.05)。结合采用脾脏/脑系数可以补充脾体系数的不足,减小了大蒜素导致体重明显降低对脾体系数的影响,更能真实反映脾脏本身的变化。[结论]大蒜素亚慢性染毒可导致大鼠脾脏和脾体系数增大。[Objective]To investigate the effect of sub-chronic exposure to allicin on spleen-body index in rats and the recovering after stopping exposure for 45d,and explore simultaneously the preliminary toxicological significance of organ-brain index.[Methods]Allicin was administered by oral garage to randomly assigned groups of rats at the dosage of 3.3 mg/kg/day,10 mg/kg/day 30 mg/kg/day respectively for continuous 90d.Rats were sacrificed,and the organ of brain,kidney and spleen were excised and weighted.The other rats were sacrificed as above after stopping exposure for 45 d.[Results]In comparison to the control rats,the weight of spleen and spleen-index of male rats intoxicated with 10mg/kg and 30 mg/kg increased relatively(P0.05 or 0.01),and the increases were observed until 45d after stopping exposure.The weight of brain was kept unaffected relatively during all the observing period(P0.05).Spleen-brain index comprised the reduced bodyweight induced by allicin,and further displayed the real changes in spleen itself.[Conclusion]Allicin sub-chronic intoxication is associated with an obvious increase in spleen index.
