三芪口服液对实验性肾炎大鼠肾小管功能及HGF、TGF-β1表达的影响

摘　　要：目的:研究益气活血为组方原则的"三芪口服液"对实验性肾炎大鼠肾小管间质的影响,并初步探讨其作用机制。方法:采用葡萄球菌肠毒素B(SEB)分次静脉注射,牛血清白蛋白(BSA)隔日灌胃、弗氏佐剂及BSA分次皮下注射,延长造模时间等方法改进大鼠系膜增生性肾小球肾炎模型。予益气活血中药复方三芪口服液治疗,观察对大鼠的病理形态学、24h尿蛋白定量、尿微量白蛋白、NAG、CysC的影响;并通过应用免疫组化、PCR等技术检测HGF、TGF-β1等细胞因子的。结果:益气活血中药复方三芪口服液能减少大鼠尿微量白蛋白、NAG酶、CysC水平;提高大鼠肾组织HGF表达,减少TGF-β1的表达。结论:益气活血中药复方三芪口服液具有肾小管功能保护作用,调节HGF、TGF-β1的表达可能是其作用机制之一。Objective：This research will investigate protective effect of Sanqi Oral Solution which is composed according to principle of Strengthening-Qi and Activating blood on renal tubulointerstitial lesion and its mechanism in mesanginal proliferative glomerulonephritis rats.Methods：With extension of modeling time,the model of mesangial glomerulonephritis with tubulointerstitial lesion were replicated by injecting staphylococcal enterotoxin B（SEB） into rat caudal vein,feeding bovine serum albumin（BSA） administered every other day,subcutaneous injection Freund＇s adjuvant and BSA periocally.After 12 and 16 weeks of treatment of Sanqi Oral Solution,the pathological morphology were observed and 24 hours urine protein,urine microalbumin,NAG and CysC were detected.Through the application of immunohistochemistryand PCR,cytokines such as HGF and TGF-β1 were detected also to explore its mechanism.Results：showed that Chinese compound formula Sanqi Oral Solution can reduce the rat urinary albumin,NAG activity,CysC level;,increase expression of renal hepatocyte growth factor（HGF） and decrease expression of TGF-β1.Conclusion：Our study showed that Chinese compound formula Sanqi Oral Solution can prevent renal tubulointerstitium injury in mesanginal proliferative glomerulonehpritis rats,which mechanism may be correlated with regulation of expressions of HGF and TGF-β1.
