摘  要:目的:通过体外实验探讨压力刺激对大鼠筋膜组织成纤维细胞形态和MMP-1、MMP-3合成释放变化的影响,为推拿基本力学刺激"舒筋散结"的治疗作用提供细胞生物学方面的实验依据。方法:体外培养大鼠筋膜组织成纤维细胞,对细胞实施100KPa和200KPa压力刺激后,观察细胞形态的变化和检测细胞MMP-1、MMP-3合成释放的变化。结果:筋膜组织成纤维细胞接受压力作用后,100KPa压力组细胞发生回缩,细胞间隙加大,200KPa压力组出现回缩成圆形并漂浮的细胞,细胞形态缩小;100KPa和200KPa压力组MMP-1合成释放呈增加趋势,但均无显著性差异(P>0.05;P>0.05);MMP-3合成释放减少,差异均有统计学意义(P<0.01;P<0.05)。结论:压力刺激对筋膜组织成纤维细胞MMP-1、MMP-3合成释放的影响和对细胞形态的影响,可能是推拿良性发挥"舒筋散结"作用的细胞生物力学原理之一。Objective:To supply experimental bases from the view of cellular biomechanics for the basical mechanical principle of massage "relieving rigidity of muscles and resolving hard mass" by exploring the research of stress on synthesis cell morphology and release of MMP-1 and MMP-3 by rat"Zusanli"fascia fibroblasts in vitro.Methods:Rat"Zusanli"Point fascia fibroblast cells were cultured in vitro.100 kPa and 200 kPa strain was taken on cells respectively.Cell morphology were observed and MMP-1 and MMP-3 of extracelluar nutrient solution were examined.Results:After being stressed,cells of group of 100Kpa strain rebound and intercellular space gets wider.While for 200Kpa group,cells rebound and become round,the volume gets smaller.The synthesis and release of MMP-1 of both groups of cells tend to increase,but the differences has no statistical significance(P0.05,P0.05).As to MMP-3,the synthesis and release increase,it has statistical significance(P0.01,P0.05).Conclusion:Pressure stimulation on regulation of synthesis and release of MMP-1 and MMP-3 of acupoints fibroblast cells and the influence on cells morphology may be one of the celluar biomechanical principles of optimum "relieving rigidity of muscles and resolving hard mass"of massage.
