摘 要:目的观察运动训练对大鼠脑缺血再灌注后不同时间神经修复及GAP-43与Neurocan表达的影响。方法健康雄性Wistar大鼠72只,随机分成运动训练组、对照组、假手术组。采用线栓法制作一侧大脑中动脉闭塞(MCAO)模型,以神经功能缺损评分和Morris水迷宫试验进行神经功能评价,免疫组化法观察对脑缺血周围GAP-43与Neuorcan的表达。结果与对照组比较,脑缺血再灌注后14d、21d,运动组的肢体运动及记忆功能明显恢复;缺血再灌注后7d,对照组缺血周围出现GAP-43阳性细胞,14d减少,21d、28d明显减少,运动组GAP-43表达在14d、21d、28d较对照组显著增加(P<0.05)。Neurocan阳性细胞在对照组缺血再灌注7d出现,14d达高峰,21d、28d时下降;运动组Neurocan表达在缺血再灌注14d、21d、28d较对照组显著减少(P<0.05)。结论运动训练上调大鼠脑缺血区GAP-43表达与下调Neurocan表达,可能是其促进脑损伤区中枢神经修复的重要机制之一。Objective To observe the effects of physical exercises on the grade of behavior and the expression of GAP-43 and Neurocan during the different time points after the cerebral ischemia and reperfusion in rat model.Methods 72 Healthy female Wistar rats were randomly divided into 3 groups:Exercising group(E group,n=24),ischemia-reperfusion group(I/R group,n=24) and sham group(S group,n=24).The middle artery cerebral occlusion(MACO) was performed with the ligation of artery with a nylon suture for 90 mins.6 rats from each group were selected to do evaluations and then sacrificed on 7d,14d,21d and 28d after MACO procedures.The evaluations included the behavior scores and Morris water maze experiment,both was to compare the recovery of neural physiological function at different time points above mentioned.The expression of GAP-43 and Neurocan were determined with immunohistochemistry(IHC) method on the tissues from all three groups.Results Compared to without exercising I/R group,the motor function and mnemonic function of exercising group were all improved on 14d and 21d after cerebral ischemia-reperfusion in exercising group.Both positive GAP-43 and Neurocan cells could be observed on 7d after reperfusion.However the positive GAP-43 cell number were significantly higher in exercising group versus without exercising group(P0.05) on 14d,21d and 28d.On the other hand,the positive Neurcan cells were significantly less in exercising group at the same time points above mentioning(P0.05).Conclusions Our data indicates that the physical exercises can improve the behavior status of rats after cerebral ischemia-reperfusion.The mechanism may be related to the up-regulation the expression of GAP-43 as well as down-regulation of the expression of Neuorcan around the ischemic areas.
