摘  要:目的探讨建立前列腺癌外周血微转移癌细胞的免疫磁珠检测方法。方法将体外培养的前列腺癌LNCaP细胞与外周血单个核细胞(PBMC)或全血混匀,与抗前列腺特异性膜抗原(PSMA)的单抗一起孵育后,经免疫磁珠分离,显微镜观察,计算玫瑰花环形成率,检测特异性和敏感性。并以抗前列腺特异性抗原(PSA)单抗作对照。结果免疫磁珠法检测前列腺癌细胞时,一抗为抗PSMA单抗优于抗PSA单抗。单个核细胞与LNCaP细胞比为5×105:1时可检测到癌细胞,每ml外周血中含20个肿瘤细胞时即可被检测出来。结论免疫磁珠法检测外周血微转移前列腺癌细胞有较高的特异性和敏感性。Aim To discuss the establishment of immunomagnetic bead-based(IMB) method for detection of circulating prostate cancer cells in peripheral blood.Methods The mixtures of prostate cancer cell line LNCaP cultured in vitro,and PBMC or all blood,were incubated with monoclonal antibody against PSMA,then isolated by using immunomagnetic beads.The isolated cells were observed through microscope to calculate the rosette formatting rates and evaluate the specificity and sensitivity of the isolate method by IMB.Results This IMB method could detect one cancer cell in 5×105 PBMC,and could detect cancer cells when there were more than 20 cancer cells in 1 ml peripheral blood.Conclusion IMB method is highly specific and sensitive in detecting prostate cancer cells in peripheral blood.
