摘 要:目的观察低剂量伽玛刀照射对青霉素点燃癫痫猫的行为和脑额叶功能区皮质神经递质的影响。方法实验于2007—2009在河北省人民医院伽玛刀治疗中心和临床研究中心完成。选择雄性成年猫80只,按随机数字表法分成3组,癫痫模型组、伽玛刀治疗组各28只,正常对照组24只。正常对照组不做任何处理;其他两组均在猫额叶功能区皮质多次注射青霉素制作点燃癫痫的模型,伽玛刀治疗组在造模成功后给予低剂量照射,DG 15Gy/50%。观察时间点为伽玛刀照射后4、8、12周。采用Smialowki 6级评分法进行行为学观察;采用高效液相色谱荧光法测定脑额叶功能区皮质谷氨酸(Glu)、γ-氨基丁酸(GABA)的含量。结果 80只猫全部进入结果分析。(1)猫行为学观察结果:除正常对照组外,各组实验用猫均出现惊厥行为,点燃率为100%。第12周时伽玛刀治疗组发作级别显著低于癫痫模型组(P<0.05)。(2)实验用猫额叶功能区皮质神经递质的含量:在各时间点,与正常对照组比较,癫痫模型组额叶功能区皮质Glu含量明显增加,GABA含量减低(P<0.05);与癫痫模型组比较,12周时,伽玛刀治疗组额叶功能区皮质Glu含量降低,GABA含量增加(P<0.05)。结论低剂量(15 Gy)伽玛刀照射对青霉素点燃癫痫猫有明显的抗癫痫作用,其机制可能与降低额叶脑功能区皮质Glu的含量和增加GABA的含量有关。Objective To observe the effect of low-dose gamma knife irradiation on both behavior and levels of neurotransmitters in the functional region of frontal cortex in kindled epileptic cat.Methods The experiment was carried out in Hebei General Hospital Gamma Knife Center and clinical research center during 2007~2009.A total of 80 male adult cats were randomized into three groups:epilepsy model group(n=28),gamma knife treatment group(n=28),and normal control group(n=24),Multiple penicillin injections were performed in the functional region of frontal cortex area to establish the kindled epileptic model.Cats in the gamma knife treatment group received a mild irradiation of DG:15Gy/50% once the model was successfully established.Animals were observed 4 weeks,8 weeks,and 12 weeks after gamma knife irradiation.Behavioral observation was conducted using the Smialowki 6-grade assessment.The levels of glutamic acid(Glu) and gamma-aminobutyric acid(GABA) in frontal cortex were determined by high performance liquid chromatography.Results Data obtained from all the 80 cats entered the final analysis.Epileptic behaviors occurred in the two modeling groups.The eclampsia grades in the gamma knife treatment group were significantly lower than those in the model group(P0.05) at week 12.Glu level in the frontal cortex was significantly higher and GABA contents were significantly lower(P0.05) in the epileptic model group than in the control group at all time points.At week 12,Glu level significantly decreased and GABA level significantly increased in the gamma knife treatment group when compared with the epileptic model group(P0.05).Conclusion Low-dose(15Gy) gamma knife irradiation has an obvious anti-eclampsia effect in penicillin-kindled epileptic cats.The mechanism may involve the decreased glutamic acid and the increased gamma-aminobutyric acid in the frontal cortex.
