KAI-1和nm23-H1的表达与结直肠癌侵袭转移的关系研究

摘　　要：目的检测结直肠癌组织中KAI-1、nm23-H1基因及蛋白的表达,探讨其在结直肠癌浸润转移中的作用及临床意义。方法以45例结直肠癌手术标本为研究对象,分别采用荧光实时定量PCR和免疫组织化学方法检测肿瘤组织和切端组织中KAI-1、nm23-H1的mRNA及蛋白表达水平,并结合临床病理资料进行统计学分析。结果结直肠癌组织中KAI-1和nm23-H1的mRNA表达水平及蛋白表达阳性率均显著低于切端组织,差异有统计学意义(P<0.01);淋巴结转移者KAI-1、nm23-H1的mRNA表达水平及蛋白表达阳性率均显著低于不伴淋巴结转移者,且二者会随着浸润深度及Dukes分期的增加而表达下降,差异有统计学意义(P<0.01)。结直肠癌组织中KAI-1 mR-NA的表达均与nm23-H1的表达呈正相关(rs值分别为0.583和0.327,P<0.05)。结论 KAI-1和nm23-H1低表达均与结直肠癌的侵袭转移有关,联合检测这两项指标有助于指导临床治疗和预测肿瘤进展。Objective To explore the relationship of KAI-1 and nm23-H1 expressions to the invasion and metastasis of colorectal carcinoma,to find their role in the invasion and metastasis.Methods The mRNA and protein levels of KAI-1 and nm23-H1 were determined by fluorescent real-time quantitative PCR and immunohistochemical methods in 45 colorectal carcinoma specimens.Results The mRNA and protein expressions of KAI-1 and nm23-H1 were significantly lower in tumor tissues than in surgical margin area（P0.01）.Expressions of KAI-1 and nm23-H1 were obviously related to lymph node metastasis,depth of invasion and Dukes staging（P0.01）.KAI-1 mRNA expression was positively correlated with nm23-H1 in colorectal carcinoma tissues（rs=0.583 and 0.327,P0.05）.Conclusion Low expressions of KAI-1 and nm23-H1 are closely associated with invasion and metastasis of colorectal carcinoma.Combined detection of the two genes may be helpful in predicting the tumor progress and guiding the clinical therapy of colorectal carcinoma.
