摘 要:目的探讨黄芪注射液抗大鼠全脑缺血再灌注损伤机制。方法采用改良的Pulsinelli四血管法,复制大鼠全脑缺血15 min再灌注7 d损伤模型,应用生化法检测海马组织中超氧化物歧化酶(SOD)、谷胱甘肽过氧化物酶(GSH-Px)、一氧化氮合酶(NOS)的活性和丙二醛(MDA)、一氧化氮(NO)的含量,并观察黄芪注射液对上述指标的影响。结果与假手术组比较,模型组海马组织SOD、GSH-Px活力下降,NOS活力明显升高;MDA、NO含量升高(P<0.05)。与模型组比较,黄芪注射液治疗组SOD、GSH-Px活力升高,NOS活力明显下降;MDA、NO含量下降(P<0.05)。结论黄芪注射液对大鼠全脑缺血再灌注损伤有保护作用,机制与提高机体抗氧化能力,抑制脂质过氧化反应,减少自由基损伤有关。Objective To investigate the action mechanism of the protective effects of Huangqi injection on cerebral ischemia/reperfusion injury in rats.Methods The cerebral schemia-reperfusion injury models of rats were established by Pulsinelli four vessels occlusion for 15min and reperfusion for 7d,and the activities of superoxide dismutase(SOD),glutathione peroxiolase(GSH-Px),nitric oxide synthase(NOS),and the contents of malondialdehyde(MDA) and nitric oxide(NO) in the hippocampal tissue of rats were detected by biochemistry,and the effect of Huangqi injection on these parameters were observed.Results As compared with those in sham-operation group,the activities of SOD,GSH-Px were decreased in model group,however,the activity of NOS was significantly increased,and the contents of MDA and NO were increased significantly in the hippocampal tissue of rats in model group(P0.05).As compared with those in model group,the activities of SOD,GSH-Px in Huangqi injection group were increased,however,the activity of NOS was significantly decreased,and the contents of MDA and No were significantly decreased(P0.05).Conclusion The Huangqi injection has protective effect on the cerebral ischemia-reperfusion injury in rats,its action mechanisms are related with enhancing anti-oxidant ability of organism,inhibiting lipid peroxidation reaction and ruducing free radical injury.
