摘  要:目的:通过免疫组化方法观察LIM-kinase 1(Limk1)在不同年龄组的FMR1基因敲除鼠(FMR1 knockout mouse,KO)和野生鼠(wild type mouse,WT)脑组织不同部位的表达差异.方法:取FVB品系新生和生后2、6周KO鼠(KO0d、KO2周和KO6周),并分别与同龄WT鼠作对照.每组6只通过免疫组化染色,观察Limk1在不同年龄组的KO和WT小鼠脑组织各部位的表达差异.结果:Limk1在新生鼠的海马、皮层、丘脑、小脑表达丰富,以染色阳性纤维为主,染色阳性细胞少见.生后2周、6周小鼠的大脑皮质和海马仅见少量散在弱阳性细胞表达,但在丘脑未定带、腹后外侧核,小脑蒲肯野细胞和脑干前庭蜗神经核可见强阳性细胞表达.各年龄组KO与WT小鼠Limk1在脑区的分布特点基本一致.KO0d、KO2周及KO6周组小脑、蜗神经核Limk1强阳性表达脑区的平均光密度值以及KO2W及KO6W组丘脑部位的阳性细胞计数均高于同龄WT组,差异有统计学意义(P<0.05).结论:Limk1在KO与WT小鼠脑组织的表达均有着明显的时空特异性,FMRP能负性调控Limk1表达.Objective:To study the difference in LIM-kinase 1 (Limkl) expression at various locations of brain tissues in FMR1 knockout mice (KO) and wild type mice (WT) in various age groups by immunohistochemistry. Methods:New born,2-week and 6-week FVB KO mice (KO0d, KO2w and KO6w) were used, with WT mice at the same ages used as controls. There were 6 mice in each group. Immunohistochemical staining was performed to observe the expression difference of Limkl at different brain tissue locations in KO and WT mice with different ages. Result:Limkl expressions were rich over hippocampus, cortex, thalamus optieus and cerebellum of newborn mice. Immunoreactive staining was basically seen in fibers but rarely in cells. In mice at 2 and 6 weeks of life, only sporadic weak immunoreactive cells were observed in cerebral cortex and hippocampus, while strongly positive expressions were found over zona incerta of thalamus, ventral posterolatera] nucleus,eerebellar Purkinje cells and brain stem auditory nucleus. The distributions of Limkl in each brain region were generally similar in KO mice and WT mice of each age group. The mean optical density of strongly positive Limkl expression region over stem and cochlear nuclei in KO0d, KO2w and NO6w groups, positive cell counts of Limkl in thalamus opticus KO2w and KO6w groups were all higher than the age-matched WT group, with statistical significance found ( P 〈 0.05 ). Conclusion :The expressions of Limkl were with obviously time series in the different brain regions of KO and WT mice. FMRP may down-regulate the expressions of Limkl.
