摘 要:选取红冠桉茎段作为外植体,建立了无菌再生体系;对外植体选择、不定芽增殖、生根诱导及移栽各阶段的关键因子进行了探讨。结果表明:(1)茎尖为较适宜的外植体;(2)最佳增殖培养基为M+6—BA 1.5 mg/L+NAA0.2 mg/L+VB26.0 mg/L;(3)最佳的生根培养基为1/2M+IBA 1.5 mg/L+VB26.0 mg/L,生根率达96.3%;(4)发酵过的椰糠与黄心土按3∶2混合而成的基质为适宜的移栽基质,移栽成活率达81.15%。本研究首次利用茎段建立了红冠桉无菌再生体系,为今后的研究及大规模生产奠定了技术基础。Stem segment of Corymbia ptychocarpa was collected as explant,and key factors for each regeneration stage,including explant selection,adventitious bud proliferation rooting induction and transplanting were tested.The results show that:(1)The stem tip was the best explant;(2) The adventitious shoots were subcultured and multiplied on an improved MS medium containing 1.5 mg/L 6-BA,0.2 mg/L NAA and 6.0 mg/L VB2;(3) Rooting medium with 1/2 MS medium supplied with 1.5 mg/L IBA and 6.0 mg/LVB2 induced a rooting rate up to 96.3%;(4) The transplant medium was the mixture of coconut husk and yellow soil by ratio of 3 to 2.The survival rate of transplants reached 81.15%.
