摘 要:目的:探讨P-gp与β-catenin和E-cadherin在结直肠癌发生发展过程中的作用及相互关系。方法:采用组织芯片、免疫组织化学技术检测135例结直肠腺癌及癌旁正常黏膜组织、89例腺瘤组织P-gp、β-catenin和E-cadherin蛋白表达情况,应用免疫荧光双重标记观察P-gp和β-catenin在腺癌组织定位情况。结果:P-gp在腺癌和癌旁正常黏膜的表达高于腺瘤(P<0.05),E-cadherin在癌旁正常黏膜和腺瘤的表达高于腺癌(P<0.05),在癌旁正常黏膜、腺瘤和腺癌组织中,β-catenin的细胞质及细胞核表达均呈逐级增高(P<0.05),P-gp与β-cate-nin细胞质/核表达与腺瘤不典型增生程度有关(P<0.05),E-cadherin表达与腺癌组织分化有关(P<0.05)。P-gp在腺瘤组织的表达与E-cadherin和β-catenin胞质表达呈正相关(P<0.05),在腺癌组织的表达与β-catenin胞质和胞核表达呈正相关(P<0.05),P-gp与β-catenin免疫荧光双重标记在腺癌组织呈共表达现象。结论:P-gp与β-catenin和E-cadherin之间存在密切联系,三者的检测对结直肠癌的早期诊断及耐药性评估有一定的帮助。OBJECTIVE:To investigate the clinical significance of P-gp,β-catenin and E-cadherin protein expression and the reciprocal relationships between them in colorectal cancer(CRC) tissues.METHODS:The expression of P-gp,β-catenin and E-cadherin protein was detected by immunohistochemistry using tissue microarrays consisting of 135 specimens of CRC and adjacent normal mucosa(ANM),89 specimens of colorectal adenoma(CRA) tissues.The co-localization of P-gp and β-catenin protein expression in CRC was detected by immunofluorescence double labeling.RESULTS:The positive rates of P-gp in CRC and ANM were higher than those in CRA(P0.05);The positive rates of E-cadherin in ANM and CRA were higher than those in CRC(P0.05);From ANM to CRA and CRC,the positive rates of β-catenin were gradually increased,both in cytoplasm and nuclear(P0.05).The expression of β-catenin in cytoplasm and nuclear were related with the dysplasia degree of CRA while E-cadherin were related with the differentiation degree of CRC(both P0.05).The expression of β-catenin in cytoplasm and E-cadherin was positively correlated with P-gp in CRA,β-catenin expression in cytoplasm and nuclear was positively correlated with P-gp in CRC(P0.05),β-catenin and P-gp was co-expressed in CRC showed by Immunofluorescence double labeling.CONCLUSION:P-gp,β-catenin and E-cadherin protein have intrinsic relationships between them,detection of these proteins may be helpful for the early diagnosis and assessment of drug resistance of CRC.
