摘  要:目的利用APP695MT基因转染PCI2细胞为细胞模型,观察植物雌激素三羟基异黄酮(GST)对PCI2细胞中Caspase-3表达的影响,初步探讨GST对表达APP695MT基因PCI2细胞的保护作用及其相关机制。方法将培养的PCI2细胞分为正常对照组、APP695转染组、GST干预组,免疫细胞化学SABC法检测凋亡相关蛋白Caspase-3在不同处理组PCI2细胞中的表达变化。,结果正常纽及空载组Pc12细胞Caspase-3阳性表达非常弱;与正常对照组比较,APP695转染组PCI2细胞Caspase-3为强阳性表达(P〈0.01);GST干预组与APP695转染组比较.Caspase-3的免疫反应产物明显减弱(P〈0.01),且15Ixmol/L和201xmol/LGST处理组与5txmol/L和10gmol/LGST处理组比较,Caspase-3的免疫反应产物减弱更明显(P〈0.01)。结论APP695基因转染的PCI2细胞中凋亡蛋白Caspase-3的表达明显增强,GST的干预能显著降低其表达,对神经细胞产生保护作用。Objective To investigate the protective effect of Genistein against apoptosis of PC12 cells induced by transfection mutant APP695 plasmid DNA and its impact on Caspase-3. Methods The AD cell model were established by transfecting the mutant APP695 plasmid DNA to PC12 cells,the expression of Caspase-3 protein in PC12 cells was obselwed by Imnumocytochemistry SABC. Results The expression level of activated Caspase-3 in normal control group and Unladen group was lower ( P 〉 0.05 ) ; Compared with the normal control group, the expression level of caspase-3 in APP695 transfected group was strongly increased(P 〈 0.01 ) ;compared with APP695 transfected group,the immune reaction of caspase-3 in all GST treatment groups were significantly decreased ( P 〈 0.01). Conclusion In APP695MT transfected PCI 2 cells,the expression level of Caspase-3 was increased,while GST could reduce the level of Caspase-3 protein in the intervention groups,which confirmed that GST has protective effects on neurons.
