摘　　要：目的观察Toll样受体4（TLR4）在血管性痴呆（VD）模型大鼠海马的表达及其动态变化，探讨TLR4介导的炎症反应在VD大鼠学习记忆障碍中的作用。方法采用双侧颈总动脉反复夹闲再灌注，同时腹腔注射硝普钠建立VD大鼠模型，在15d、1个月、2个月和4个月等时间点，采用Morris水迷宫试验检测假手术组和模型组大鼠学习记忆能力的变化，免疫荧光技术观察两组大鼠海马TI，R4形态、数量及表达的变化。结果与假手术组相比，模型组大鼠EL均明显延长（P〈0．01）．且模型组各时间点EL随时间的延长而逐渐延长。模型组大鼠海马TLR4表达比假手术组明显增多；组问比较，随造模手术后时间的延长，15d和1个月时间点间TLR4阳性细胞在海马CA1区逐渐增加，2个月和4个月时间点间则在海马CAI区和DG区均明显增多趋势。结论TLR4介导的炎症反应对VD大鼠长时间海马损伤发挥重要作用，对其学习记忆功能障碍的发生发展也可能起到重要作用。Objective To observe the expressions and dynamic changes of TLR4 in hippocampus of vascular dementia（VD） rats. Methods VD rat models were established by repeatedly cerebral ischemia-reperfusion. Mort＇is water maze was used to detect the changes of spatial learning and memol＇y abilities. The changes of morphology and the amount of TLR4 expression in hippocampus were observed by immunofluorescence in both sham-operated（SO） group and VD model group on 15th d, I st month,2nd month and 4th month. Results （1）The escape latency（EL） of model group was obviously longer than that of SO group on different time points（ P 〈 0.01 ）. （2）The expression amount of TLR4-positive substance in hippocampus of Model group was significantly increased compared with SO group. （3）Compared with SO group, TLR4 positive expression were mainly found in hippocampal CAI region of model group on 15th d and 1st month;and an significantly increased expression level of TLR4 was observed in both hippocampal CA1 region and DG area of model group on 2nd month and 4th month. Conclusion TLR4-mediated inflammatory reaction plays an important role in long-term hippocampal damage of vascular dementia, which may be one of the mechanisms in learning and memory, ability dysfunction of vascular dementia.
