The Effect of Human IL-23 on the Secretion of Human IL-17 in the Peripheral Blood Mononuclear Cells of Ulcerative Colitis Patients

摘  要:观察人白介素-23(Interlerleukin,IL-23)刺激对溃疡性结肠炎(ulcerative colitis,UC)、感染性腹泻患者及健康对照者的外周血单个核细胞(peripheral blood mononuclear cells,PBMCs)分泌人白介素-17(Interlerleukin,IL-17)的影响。分离、培养17例UC患者、10例感染性腹泻、13例健康对照者的PBMCs;采用ELISA方法检测入组者PBMCs经IL-23刺激前、后的培养上清液中IL-17水平。未经IL-23刺激的PBMCs培养上清液中IL-17水平在UC组为(29.49±6.63)pg/mL、感染性腹泻组(18.18±3.11)pg/mL、健康对照组(16.85±3.90)pg/mL,UC组高于其他两组(P<0.05)。经IL-23刺激的PBMCs培养上清液中IL-17水平在UC组为(63.63±11.79)pg/mL,较感染性腹泻组(7.87±8.31)pg/mL和健康对照组(26.53±6.42)pg/mL的升高幅度更为明显(P<0.05)。结果提示IL-23可能具有促进PBMCs分泌IL-17的作用。IL-23可能通过诱导IL-17分泌而使后者在UC的慢性病程中发挥重要作用,IL-23/IL-17免疫通路失调而产生的对机体免疫耐受机制的破坏作用,使其成为导致UC发生的原因之一。The effect of human IL-23 on secretion of human IL-17 in the peripheral blood mononuclear cells(PBMCs) of ulcerative colitis patients(UC) 、Infectious diarrhea patients and healthy controls is observed.PBMCs(peripheral blood mononuclear cells) are isolated and cultured from 17 cases of UC,10 cases of Infectious diarrhea patients and 13 healthy controls.The amounts of human IL-17 induced or not by human IL-23 in supernatants of cultured PBMCs from each group are measured by ELISA.The amounts of human IL-17 which were not induced by human IL-23 in ulcerative colitis patients(UC) were(29.49 ± 6.63) pg/mL,the infectious diarrhea patients(18.18 ± 3.11) pg/mL and the healthy controls(16.85 ± 3.90) pg/mL respectively.UC group are higher than other groups(P0.05).The amounts of human IL-17 which are induced by human IL-23 in ulcerative colitis patients(UC) were(63.63±11.79) pg/mL,the infectious diarrhea patients(27.87±8.31) pg/mL and the healthy controls(26.53±6.42) pg/mL respectively.The PBMCs from all groups,and that of UC group is significantly higher than that of other two groups(P0.05).The results show the human IL-23 might stimulate the production of human IL-17.The human IL-23 might play a critical role in the development of UC through inducing production of human IL-17.Impaired human IL-23/IL-17 path way may induce UC by destructing immune tolerance.
