摘  要:本文将极限稀释微孔培养CFU-GM技术初步用于急性非淋巴细胞白血病(ANLL)和慢性粒细胞白血病(CML)研究中,对两者的CFU-GM进行了细胞形态学、细胞化学染色及生化标记、扫描电镜、~3H-TdR掺入和细胞遗传学等方面的研究,证实极限稀释微孔培养较琼脂半固体培养更有利于白血病克隆祖细胞的生长。In vitro granulocyte-macrophage progenitor (CFU-GM) with limiting dilution microculture was preliminarily used in the study of acute and chronic myelogeneous leukemia (AML and CML).It was proved that CFU-GM in AML and CML were generated more suitably in the limiting dilution microculture than in semisolid agarculture by the studies of CFU-GM with cytomorphology,cloning efficiency,cytochemistry,scanning electron microscopy,3H-TdR uptake and cytogenetics.In addition,four different growth patterns were observed in vitro.
