摘　　要：目的:应用RNAi技术封闭Survivin基因,对甲状腺癌细胞增殖的影响,探讨RNA干扰survivin基因表达治疗甲状腺癌的可行性。方法:实验设为空白组、脂质体组、阴性对照组和RNAi组。RNAi组是针对Survivin基因的有效序列合成DNA正义链及反义链,经变性、退火,形成双链DNA,与pGenesil-1线性质粒构建重组体,经脂质体转染SW579细胞。各组利用RT-PCR、Western-bloting检测survivin表达情况;MTT法检测细胞增殖的变化,流式细胞仪检测转染后SW579细胞凋亡的变化。结果:RT-PCR、Western blotin结果显示,转染survivin基因重组体组的mRNA和蛋白水平的表达与其它各组比较均明显下调(P<0.01),转染后SW579细胞生长率下降,凋亡指数明显增加(P<0.01)。结论:特异封闭survivin基因,能够有效地抑制甲状腺癌细胞的增殖,诱导其凋亡的发生。Objective：To observe effect of inhibiting survivin gene by RNA interference on thyroid carcinoma SW579 cell line in growth and apoptosis,in order to investigate feasibility of treatment on thyroid carcinoma.Methods：Cells were divided into control group,liposome group,negative group and RNAi group.RNAi group was done that target gene segments were synthesized and cloned into the vector,to construct recombinant eukaryotic expression vectors.Then the SW579 cells were tranfected with recombinant vectors.Interfering effects were detected by RT-PCR and Western blotting.The proliferation of the SW579 cells were measured by MTT method.Apoptosis index were detected by flow cytometry.Results：The results of RT-PCR and Western bloting indicated that knocking down expression of survivin gene Obviously decrease in growth of the SW579 cells（P0.01）.Apoptosis of index increased（P0.01）.Conclusions： Knocking survivin gene can effectly inhibit the growth of SW579 cells,and promote apoptosis of SW579 cells.
