摘  要:以近缘种OPN的cDNA序列为参考设计引物,通过RT-PCR得到黑线姬鼠OPN部分序列,并将得到的cDNA序列翻译成氨基酸序列,注册到GenBank(登陆号:HM626723).利用该序列构建系统进化树,同时对其编码的蛋白质进行一级结构分析及二级结构预测.结果表明:克隆得到的209 bp OPN的序列,包括部分外显子4,全部的外显子5及部分外显子6;该段序列从第二个碱基开始翻译,共编码69个氨基酸;其一级结构中包括了一个由10个连续天冬氨酸组成的高度酸性区域,它对骨桥蛋白与羟基磷灰石之间的联系具有重要意义;二级结构中大部分为α螺旋,且具有亲水性;利用所得OPN序列构建的系统进化树显示黑线姬鼠与大鼠、小鼠亲缘关系最近,与鸡、鹌鹑亲缘关系最远,此结果与物种的进化关系相一致,本研究所得到的OPN序列将有助于对骨桥蛋白的作用机制研究及系统进行分析.Partial cDNA sequence of the Osteopontin was cloned by designed primer and RT-PCR method.The cDNA sequences were translated into amino acid sequence and were signed them into GenBank(accession number:HM626723).The phylogentic trees were constructed based on OPN cDNA sequences;the primary secondary structure of the protein were analyzed by bioinformatics.Results shown that: 69 amino acids were translated from the 209 bp partial OPN which include partial exon 4,the whole exon 5 and partial exon 6;there is a highly acidic region composed of 10 aspartates existed in OPN primary structure,and they were most likely responsible for the binding of OPN to hydroxyapatite;α helix occupied higher proportion in secondary structure but there was no β fold,and behaved well hydrophilic;the phylogentic trees showed that the relationships between Apodemus agrarius and Rattus norvegicus,Mus musculus were the closest,but away from Gallus gallus and Coturnix coturnix,which is consistent with evolutionay classes of certain species,therefore this OPN sequence has important referential value for study on mechanism and phyletic evolution of osteopontin.
