羊栖菜褐藻糖胶的抗凝血和促血管内皮细胞生长活性研究

摘　　要：采用乙醇分级沉淀法对酸提法从羊栖菜中提取的粗糖进行纯化得到褐藻糖胶。采用体外凝血试验tAPTTt、TTt、PT评价了褐藻糖胶体外抗凝血活性和MTT染色法检测褐藻糖胶对人脐静脉血管内皮细胞(HUVEC)生长的促进作用。结果表明t,APTTt、TT分别在不同浓度与对照组出现显著差异,而tPT未出现显著差异。在10μg/mL～200μg/mL浓度范围内,人脐静脉血管内皮细胞增殖数量有大幅度提高,对人脐静脉血管内皮细胞增殖具有明显促进作用,说明褐藻糖胶是促人脐静脉血管内皮细胞生长的活性物质。Fucoidan from Sargassum fusiforme Setchel was extracted by acid method and purified by fractional precipitation with alcohol.Its anticoagulant activities in vitro and cell proliferation of Human umbilical vein endothelial cells（HUVEC） were appreciated preliminarily by the method of activited partial thromboplastin time（tAPTT）,activited thromboplastin time（tPT）,thrombin time（tTT） and method MTT staining respectively.It is shown that tAPTT,tTT have Significantly different from the control in different concentration respectively,and tPT has not.The cell proliferation of HUVEC raises more in range of 10 μg/mL-200 μg/mL,It could promote the growth and proliferation of HUVECs significantly.There are some components of promotion growth of HUVECs in the fraction from Sargassum fusiforme Setchel.
