摘 要:目的:探讨抗癌活性肽(ACBP)对小鼠肝癌H22移植瘤生长的抑制作用及其可能机制。方法:建立小鼠H22肝癌细胞实体瘤模型,并将其随机分为三组,分别给予5-Fu(20 mg/kg),ACBP(0.4 mL/只)和生理盐水(0.4 mL/只),尾静脉注射,隔日给药,连续10 d;然后处死小鼠,计算重量抑瘤率(IR)和脾指数(SI),并用流式细胞仪检测细胞周期和凋亡。结果:ACBP能显著抑制肿瘤生长,IR最高达54.20%;流式细胞术分析结果显示ACBP能阻滞肿瘤细胞于G0/G1期,并能促进瘤细胞凋亡。结论:ACBP对小鼠肝癌有显著的抑制作用,其作用机制可能是通过诱导肿瘤细胞凋亡,抑制DNA合成,从而达到抗肿瘤效应。Objective:To investigate the effects of anti-cancer bioactive peptide(ACBP) on hepatocarcinoma H22 tumor-bearing mice,and to elucidate the possible mechanisms of inhibition.Methods: Animal model of hepatocarcinoma H22 tumor-bearing mice were established.And the mice were randomly divided into three groups,and were treated with 5-Fu,ACBP,physiological saline by caudal vein injection,respectively.The mice were fed 10 days and were treated every other day.Then the mice were executed,inhibitory rate(IR) and spleen index(SI) were calculated.Cell cycle and apoptosis were detected by flow cytometry.Results: Tumor growth was inhibited by ACBP.The IR was 54.20%.Cell cycle was stopped at the G0/G1 phase and apoptosis was induced by ACBP.Conclusion: ACBP could inhibit DNA synthesis of liver cancer cells and induce apoptosis
