摘  要:建立反相高效液相色谱法同时测定大籽獐牙莱中10种成分的含量。方法:采用反相高效液相色谱,色谱条件为:AgilentXDB C18柱(中4.6×150mm,5mm);流动相A:甲醇,B:水;洗脱方法:0~20min,A为10%~30%,20~45minA为30%~80%梯度洗脱;体积流量1.0mL/min,检测波长254nm,230nm,柱温28℃。结果:10种成分均达到基线分离,各化合物的线性关系良好,r〉0.9995;各待测物的精密度RSD值均小于1.1%;重复性良好(RSD〈3.7%);回收率范围在97.8%~104.2%,RSD〈C4.2%。结论:该方法测定成分多,结果准确、可靠。Objective:To establish a RP-HPLC method for simultaneous determination of 10 chemical components in large Swertia seeds.Methods: RP-HPLC was used,with chromatographic conditions: agilent XDB C18 column(Φ4.6×150 mm,5 μm);mobile phase A: methanol,B: water;elution: 0~20 min;gradient elution,A: 10%~30%,20~45 min A: 30% to 80%;volume flow rate 1.0 mL/min,detection wavelength 254 nm,230 nm;column temperature 28 ℃.Results: 10 components were baseline separated,with a good linear relationship with the compounds,r>0.999 5;the precision of the analytes was less than 1.1% RSD,with a good reproducibility(RSD<3.7%);recovery at the range of 97.8%~104.2%,RSD<4.2%.Conclusion: The method can determine more components and the results are accurate and reliable.
