摘 要:目的:观察新生大鼠缺氧缺血后组织型转谷氨酰胺酶(TG-2)与N-ε-γ谷氨酰基赖氨酸交联物(GGEL)在海马CA1区的表达特点,探讨TG-2与GGEL是否参与新生鼠缺氧缺血性脑损伤(HIBD)过程。方法:新生7日龄SD大鼠80只,随机分为对照组和实验组,每组40只。实验组在乙醚麻醉下行右侧颈总动脉结扎,氮氧混合气体(92%氮气和8%氧气)缺氧2 h,制作为HIBD模型。对照组仅分离颈总动脉,不结扎,不做缺氧处理。分别于缺氧后不同时间点(1天、2天、3天、5天、7天)断头,取脑组织。应用免疫组化法检测TG-2与GGEL的表达情况。结果:①免疫组化显示海马CA1区对照组各个时间点均有TG-2的表达,且没有表达量的变化,实验组海马区TG-2在缺氧缺血(HI)后3天开始表达逐渐增加,5天达到表达高峰,随后表达有所下降(P<0.05)。②海马CA1区对照组各时间点有GGEL的阳性细胞表达,但量少,且比较没有统计学差异,实验组HI后1天起GGEL的阳性细胞数表达较对照组开始增加,于HI后5天达到表达高峰,较对照组差异有统计学意义(P<0.05)。结论:TG-2与GGEL在缺氧缺血性脑损伤后的表达变化提示TG-2和GGEL可能参与了新生鼠缺氧缺血性脑损伤过程。Objective:To observe the expression characteristics of tissue transglutaminase(TG-2) and GGEL in CA1 region of hippocampus of neonatal rats after hypoxia and ischemia,explore whether TG-2 and GGEL participate in the course of hypoxic-ischemic brain damage(HIBD).Methods:80 neonatal SD rats aged 7 days old were divided into control group and experimental group randomly,40 rats in each group.The rats in experimental group received right common carotid artery ligation after anesthetized with ethylether,then the models of HIBD were established after oxygen deficit for 2 hours(92% nitrogen and 8% oxygen).The rats in control group received right common carotid artery exclusion,but right common carotid artery ligation and oxygen deficit were not carried out.The brain tissues of the two groups were obtained at 1,2,3,5 and 7 days after oxygen hypoxia,immunohistochemical method was used to detect the expression levels of TG-2 and GGEL.Results:Immunohistochemistry showed that TG-2 expressed in CA1 region of hippocampus at all time points in control group,but there was no change in expression level of TG-2;in experimental group,the expression level of TG-2 in CA1 region of hippocampus increased gradually on the third day after hypoxia,peaked on the fifth day,then decreased(P0.05).The positive cells of GGEL expressed in CA1 region of hippocampus at all time points in control group,but the expression level was low,and there was no significant difference;in experimental group,the number of positive cells of GGEL in CA1 region of hippocampus increased on the first day after hypoxia,peaked on the fifth day,there was significant difference between the two groups(P0.05).Conclusion:The changes of TG-2 and GGEL expressions after HIBD indicate that TG-2 and GGEL may be involved in the course of HIBD in neonatal rats.
