胃癌中NF-κBp65、Caspase8的表达及临床意义

摘　　要：目的:探讨NF-κBp65、Caspase8在胃癌中的表达及与胃癌临床病理特征之间的关系。方法:采用免疫组化PV-9000二步法检测60例胃癌组织标本中NF-κBp65、Caspase8两种蛋白的表达。结果:胃癌组织中NF-κBp65阳性表达率为63.3%,明显高于癌旁胃组织的30.0%P<0.05),Caspase8在胃癌中的阳性表达率为35.0%,明显低于癌旁胃组织的83.3%(P<0.05);胃癌组织分化程度越低,NF-κBp65阳性表达率越高,而caspase8阳性表达率越低P<0.05);NF-κBp65的表达率随肿瘤的浸润深度增加而增高,有淋巴结转移者比无转移高(P<0.05),而caspase8的表达与肿瘤的浸润深度和淋巴结转移均无明显关系(P>0.05);二者在胃癌中的表达呈负相关(P<0.05)。结论:NF-κBp65、Caspase8通过调节细胞凋亡,参与胃癌的发生发展过程,且在胃癌中NF-κBp65能够下调Caspase8的表达。Objective：To investigate the protein expressions of NF-κBp 65 and caspase 8 in gastric carcinoma tissues and their relationship with clinicolpathological features.Methods：NF-κBp65 protein and caspase 8 protein were determined in 60 cases of gastric carcinoma by immunohistochemistry PV-9000.Results：The positive rate of NF-κBp65 protein expression was 63.3% in gastric carcinoma tissues,higher than 30.0% in adjacent non-cancerous tissues（P0.05）,the positive rate of caspase 8 protein expression was 35.0% in gastric carcinoma tissues,lower than 83.3% in adjacent non-cancerous tissues（P0.05）;the expression of NF-κBp65 in poorly-differentiated adenocarcinoma was higher than that in well-differentiated adenocarcinoma（P0.05）,but the expression of caspase 8 in poorly-differentiated adenocarcinoma was lower than that in well-differentiated adenocarcinoma（P0.05）;and the increased expression of NF-κBp65 significantly correlated with infiltrative depth,lymph node metastasis（P0.05）.The expression level of caspase 8 only was correlated with differentiation of gastric carcinoma;a negative correlation also was founded between NF-κBp 65 and caspase 8（P0.05）.Conclusion：NF-κBp65 and caspase 8 may be involved in apotosis of gastric carcinoma and play an essential role in carcinogenesis.NF-κBp65 may have negative regulation on caspase8 in gastric carcinoma.
