马齿苋水煎剂对溃疡性结肠炎小鼠结肠组织中IL-8、IL-10表达的影响

摘　　要：目的:探讨马齿苋水煎剂对溃疡性结肠炎(UC)模型小鼠的作用机制。方法:清洁级雄性昆明小鼠给予自由饮用3%的葡聚糖硫酸钠溶液(DSS)10天,以制备UC模型,分别设空白组、模型组、柳氮磺胺吡啶肠溶片组(西药组)、马齿苋水煎剂灌胃组(中胃组)、马齿苋水煎剂灌肠组(中肠组)、马齿苋水煎剂灌胃联合灌肠组(中胃肠组),给药14天,分别于造模后、给药7天、给药14天时分批处死小鼠,取结肠组织,用酶联免疫法(ELISA法)检测结肠组织匀浆中IL-8、IL-10的含量。结果:马齿苋水煎剂灌胃联合灌肠对小鼠结肠组织中IL-10的提高和IL-8的降低均能取得与西药柳氮磺胺吡啶肠溶片相当的作用。结论:马齿苋水煎剂明显调节了UC小鼠结肠组织中促炎因子与抗炎因子之间的不平衡,下调了局部免疫反应状态,减轻了黏膜炎症损伤,这些作用可能是其治疗溃疡性结肠炎的机制之一。Objetive：Purslane water decoction of ulcerative colitis（UC）model in mice and its mechanism.Methods：Clean male Kunming mice were given free drink 3% solution of dextran sulfate sodium（DSS）10 days to prepare UC models,respectively,a blank group,model group,sulfasalazine enteric-coated tablets group（Western medicine group）,purslane decoction fed group（in the stomach group）,Portulaca oleracea decoction enema group（in the intestinal group）,Portulaca oleracea decoction enema group fed the joint（in the gastrointestinal group）administration for 14 days,respectively,after modeling,administration of 7 days,mice were killed 14 days after administration,colonic tissue obtained by enzyme-linked immunosorbent assay（ELISA）detection of colon tissue in IL-8,IL-10 content.Results：The decoction of fed purslane United enema colon tissues of mice increased IL-10 and IL-8 can reduce the acquisition and Western Sulfasalazine enteric-coated tablets considerable role.Conclusion：The significant adjustment of the water decoction of purslane UC mouse colon tissue factor pro-inflammatory cytokines and anti-inflammatory imbalance between the reduced state of the local immune response,thereby reducing the mucosal inflammatory damage,these effects may be the treatment of ulcers one of the mechanisms of colitis.
