摘 要:目的探讨灯盏花素保护大鼠肾脏缺血-再灌注(I-R)损伤的机制。方法 SD大鼠30只,分为假手术对照(Sham)组,I-R组和灯盏花素组。免疫组织化学法检测肾脏L-选择素蛋白的表达。检测血清、肾组织一氧化氮(NO)、一氧化氮合酶(NOS)、超氧化物歧化酶(SOD)和丙二醛(MDA)水平。结果与Sham组相比,I-R组血清、肾组织MDA明显升高,SOD明显降低(P均<0.05);灯盏花素组血清、肾组织MDA低于I-R组,SOD高于I-R组(P均<0.05)。I-R组血清和肾组织NO、NOS明显高于Sham组,灯盏花素组NO、NOS水平低于I-R组(P均<0.05)。I-R组肾组织L-选择素蛋白表达明显高于Sham组,灯盏花素组L-选择素蛋白表达低于I-R组。结论灯盏花素对肾脏有保护作用。可能机制与其减少L-选择素蛋白表达,提高SOD活性,降低NOS、NO、MDA水平有关。Objective To investigate the mechanism of breviscapine in protecing the kidney from ischemia-reperfusion(I-R) injury in rats.Methods Thirty SD rats were divided into three groups with 10 rats each.Group C was sham operated,group I-R experienced ischemia for 1h and reperfusion for 2h,and group B was given breviscapine 6 mg/kg at 30 min before operation.L-selectin protein expression was detected by immunohistochemical method.The contents of malonialdehyde(MDA) and nitric oxide(NO) in serum and kidney tissues were measured.The activities of superoxide dismutase(SOD) and nitric oxide synthase(NOS) in serum and kidney tissues were measured as well.ResultsCompared to group C,the contents of MAD in serum and kidney tissues in group I-R were higher,but those of SOD were lower(P0.05).The contents of SOD in serum and kidney tissues were higher in group I-R than those in group B(P0.05),but the contents of MDA were lower in group B than those in group I-R(P0.05).The contents of NO and NOS in serum and kidney tissues were higher in group I-R than those in group C(P0.05),which were lower in group B than those in group I-R(P0.05).The expression of L-selectin in kidney tissues was higher in group I-R than that in group C(P0.05),which was lower in group B than that in group I-R(P0.05).Conclusion Breviscapine has obvious protective effects on renal I-R injury.The mechanism probably is associated with reducing L-selectin protein expression,decreasing the level of NO,NOS,MDA and increasing the level of SOD in serum and the kidney.
