摘 要:利用复凝聚法制备了粒径约3.0 mm的羧甲基纤维素-壳聚糖胶囊,并用其固定化蔗糖酶。羧甲基纤维素-壳聚糖胶囊固定化蔗糖酶的最佳制备工艺条件是:蔗糖酶溶于质量分数为0.02的壳聚糖溶液,滴入质量分数为0.015羧甲基纤维素溶液中并在其中固化24 h。固定化酶耐酸性好,在pH3.6~5.0范围内其活性基本保持不变,而游离酶在pH3.6时其活性仅为最大时的63%;固定化酶的米氏常数(Km=54.11 mmol/L)大于游离酶的米氏常数(K=35.62 mmol/L)。3.0 mm diameter chitosan-sodium carboxymethyl cellulose(CMC) capsule was made by complex coacervation method and it was used to immobilize sucrase.The optimal immobilization conditions were: the sucrase was dissolved in chitosan solution with the concentration of 0.02.The solution was dropped into the CMC solution with concentration of 0.015,and the formed capsules were kept hardening for 24 h at room temperature.The immobilized enzyme was more stable to pH values.The enzyme activity of the immobilized sucrase had adaptive capacity at pH 3.6~5.0,while the free enzyme had only 63% relative activity at pH 3.6.The Km of immobilized sucrase was 54.11 mmol/L,higher than that of free sucrase(35.62 mmol/L).
