摘　　要：目的探讨5-氮杂-2'-脱氧胞苷(5-Aza-CdR)对胃癌细胞系SGC7901的p27kip1基因异常甲基化及表达的影响。方法选择不同浓度的5-Aza-CdR处理体外培养的SGC7901细胞后,用MSP法检测用药前后细胞中p27kip1基因的甲基化状态;RT-PCR法检测用药前后细胞中p27kip1的mRNA及蛋白表达的变化。结果 p27kip1基因的甲基化状态得到了逆转,p27kip1基因的mRNA和蛋白表达得到增加。结论 SGC7901细胞中p27kip1基因的表达情况与其甲基化状态的改变有关。Objective To study the effect of 5-Aza-CdR on p27kip1 gene＇s abnormal methylation in gastric cancer cell line.Methods The cells were treated with different concentrations of 5-Aza-CdR.The methylation of p27kip1 in the cell line was detected by MSP,and the expression of p27kip1 was detected by RT-PCR and westernblot.Results The methylation of p27kip1 cells were changed with 5-Aza-CdR.The expression of p27kip1 mRNA and protein increased.Conclusion The p27kip1 gene＇s expression is related with the gene＇s methylation states.
