胃癌组织端粒酶活性表达与临床病理特征相关性的研究

摘　　要：目的探讨胃癌组织端粒酶活性表达与其临床病理特征的关系。方法应用端粒酶重复序列扩增(telom-eric repeat amplification protocol,TRAP)银染TRAP-ELISA方法检测25例胃癌组织(实验组)和25例胃不典型增生组织(对照组1)及正常胃组织(对照组Ⅱ)中端粒酶活性的表达,并胃癌组织与临床病理分期、病理分级及预后关系进行分析。结果实验组中端粒酶活性表达率为91.1%,对照组Ⅰ端粒酶活性表达率为20%,对照组Ⅱ无表达,实验组端粒酶活性高于两对照组(P<0.01);实验组端粒酶活性在临床病理分期上:T4高于T3,T3高于T2+T1,但差异均无显著性。实验组端粒酶活性在病理分化程度上:未分化高于高分化(P<0.05)。结论实验组端粒酶活性高表达,与胃癌临床病理分化程度呈正相关,端粒酶激活可能在胃癌的发生及发展过程中起重要作用,端粒酶可作为胃癌早期诊断及判定预后的分子生物学标记。Objective To discuss the relationship of telomerase activity and the clinical characteristic in gastric carcinoma.Methods Telomerase activity was detected by telomeric repeat amplification protocol（TRAP）-Ag stain and ELISA in 25cases of gastric carcinoma（as experiment group）and 25 cases of gastric atypical hyperplasia tissues（as control group） The relationship of telomerase activity and the clinical stage,pathologic grades and prognosis were assayed in gastric carcinoma.Results The positive rate of telomerase activity was 91.1% in experiment group and 20%control groupⅠand 0 in control groupⅡ respectively.The telomerase activity in experiment group was higher than that in control group（P0.01）.Telomerase activity in T2 was higher than that in T1,T3 was higher than that in T2.But there were not significant difference.Telomerase activity in Histological type there were significant difference.（P0.05）.Conclusion Telomerase activity overexpressions in gastric carcinoma could be positively related to the clinical pathologicgrades.Telomerase activation could play an important role in the tumor genesis and progress in,gastric carcinoma which could serve as molecular biology marker of early diagnosis and prognosis judgment.
