摘 要:目的观察灵五颗粒对刀豆蛋白A所致小鼠免疫性肝损伤的保护作用。方法采用尾静脉注射刀豆蛋白A致小鼠肝损伤模型。60只小鼠随机分为正常组、刀豆蛋白A模型组、灵五颗粒低、中、高(2.5、5、10 g/kg)剂量组和阳性药联苯双酯(0.15 g/kg)组。各组动物连续灌胃给药5、7 d后,尾静脉注射刀豆蛋白A(20 mg/kg)。实验结束测定各组动物血清中ALT、AST等指标;称取肝、脾、胸腺重量,计算脏器指数;检测肝组织中MDA、SOD、NO、GSH的活性变化,并观察肝脏病理学改变。结果灵五颗粒各剂量组能明显减轻肝、脾肿大,增大胸腺重量,尤以高剂量组最为明显;显著降低小鼠血清中ALT、AST活性,降低肝内MDA、NO含量和升高SOD、GSH活性;病理结果表明,与模型组比较,灵五颗粒中、高剂量治疗组能明显减轻免疫性小鼠肝损伤的损伤程度。结论灵五颗粒对刀豆蛋白A诱导小鼠免疫性肝损伤具有较好的保护作用,有明显降酶效果,其作用机制可能与其抗氧化损伤有关。Objective To study the protective effect of Lingwu Granula on immunological liver injury induced by concanavalin A in mice.Methods A model of immunological liver injury was induced by concanavalin A in mice.60 mice were randomly divided into six groups: normal,model,Lingwu Granula treatment low,medium,high(2.5,5,10 g/kg) and bifendate group(0.15 g/kg).After intragastric administration of corresponding drugs for 5 and 7 consecutive days,mice were injected by vena caudalis with concanavalin A(20 mg/kg).The activity of ALT,AST in serum was detected.The weight of the liver,spleen,thymic and its organ index were calculated.The change of activities of MDA,SOD,NO and GSH in the liver tissue was detected.In addition,hepatic histopathology was observed.Results Each group of Lingwu Granula could reduce the sweling of the liver and spleen,increase thymic weight,especially for the high dosage group of Lingwu Granula.High dosage group of Lingwu Granula could also decrease the content of ALT,AST in serum,the activities of MDA,NO in the liver and increase the activities of SOD,GSH.The pathological results showed that the medium,high group of Lingwu Granula could alleviate liver injury compared with model group.Conclusion Lingwu Granula has protective effect on immunological liver injury in mice induced by concanavalin A.The mechanism may be related to its antioxidant activity.
