摘 要:目的:比较改变血红素加氧酶-1(HO-1)水平对内毒素血症大鼠胸主动脉和肺动脉血管舒缩功能的影响。方法:将SD大鼠随机分为对照组和内毒素模型组(LPS4mg/kg,腹腔注射)。应用RT-PCR技术检测血管HO-1mRNA表达;应用血管张力检测技术检测各组主动脉和肺动脉血管环在HO-1抑制剂和诱导剂孵育前后反应性变化。结果:内毒素组胸主动脉与肺动脉的HO-1mRNA均明显增高。HO-1抑制剂预孵育后,内毒素组胸主动脉对苯肾上腺素(PE)的收缩反应较孵育前提高了31.42%(P<0.01),但对乙酰胆碱的舒张反应无显著变化;内毒素组肺动脉对PE的收缩值亦增加(P<0.01)。HO-1诱导剂预孵育后,内毒素组胸主动脉收缩性较孵育前显著下降,肺动脉收缩性较孵育前虽有下降趋势,但差异无统计学意义。结论:HO-1对内毒素血症胸主动脉和肺动脉的舒缩功能的影响存在差异。Objective To compare the effects of heme oxygenase-1 (HO-1) on the vasomotor function of thoracic aorta and pulmonary artery in rats with endotoxemia. Methods Sprague-Dawley rats were randomly assigned into control group and cndotoxemia group (LPS 4 mg/kg, i. p). The expression of HO-1 mRNA was detected by RT-PCR. Reactivity of aortic rings was detected by the isolated artery ring technique. Results The expression of HO-1 mRNA was increased in both artery in endotoxemia group. After being incubated with HO-1 inhibitor, the contractile response to PE was increased by 31.42% in thoracic aortic rings in endotoxemia group, but the relaxation response to acetylcholine was not affected; the contractile response to PE in pulmonary artery rings was increased too. The contractile response of thoracic aortic rings in endotoxemia group was decreased after being incubated with HO-1 inducer for 20 minutes. Conclusions There are significant differences between the effects of HO-1 on the vasomotor function of thoracic aorta and pulmonary artery rats with endotoxemia.
