摘 要:目的研究普罗帕酮对豚鼠左心室流出道自律细胞电活动的影响以及普罗帕酮对肾上腺素(a-drenaline,Adr)所致该部位自律性改变的影响。方法采用常规玻璃微电极细胞内电位记录技术,分别记录并分析了普罗帕酮对豚鼠左心室流出道自发慢反应电位的影响,以及普罗帕酮对Adr所致该电位改变的影响。观测指标有:4相自动除极速度(VDD)、自发放电频率(RPF)、最大舒张电位(MDP)、0相最大除极速度(Vmax)、动作电位幅度(APA)以及复极50%和80%时间(APD50 and APD80)。结果①0.5μmol/L普罗帕酮可使左心室流出道自发慢反应电位VDD和RPF减慢,Vmax减慢,APD80延长(P<0.05)。②1μmol/L普罗帕酮可使该自发慢反应电位RPF减慢,MDP绝对值和APA减小,Vmax减慢,APD50延长(P<0.05),VDD减慢和APD80延长(P<0.01)。③10μmol/L普罗帕酮可使该自发慢反应电位VDD和RPF显著减慢,APA减小,APD80延长(P<0.01),MDP绝对值减小,Vmax减慢,APD50延长(P<0.05)。④用10μmol/L Adr灌流标本10 min,VDD和RPF明显加快,MDP绝对值和APA明显增大,Vmax加快,APD50显著缩短(P<0.01);随后用1μmol/L普罗帕酮+10μmol/LAdr灌流,和10μmol/L Adr灌流组相比VDD明显减慢(P<0.01),RPF明显减慢,MDP绝对值和APA减小,Vmax减慢,APD50和APD80延长(P<0.05)。结论普罗帕酮可降低左心室流出道自律细胞的电活动,同时对Adr导致的该部位自律性异常有一定的影响。【Objectives】 To investigate the electrophysiological effects of Propafenone on autorhymic cells in ventricular outflow tract and the effects of Propafenone on adrenaline(Adr).【Methods】 By using conventional intracellular microelectrode technique to record action potentials,Propafenone was used to observe the effects on spontaneous action potentials of left ventricular outflow tract.The effects of Propafenone on Adr were also studied.Electrophysiological parameters examined are: velocity of diastolic depolarization(VDD),rate of pacemaker firing(RPF),maximal diastolic potential(MDP),maximal rate of depolarization(Vmax),amplitude of action potential(APA),50% and 80% of duration of action potential(APD50 and APD80).【Results】 0.5 μmol/L Propafenone resulted in a significant decrease in VDD and RPF,a notable decrease in Vmax,a marked lengthening in APD80(P 0.05).1 μmol/L Propafenone resulted in a significant decrease in VDD(P 0.01) and RPF(P 0.05),a notable decrease in MDP,APA and Vmax(P 0.05),a marked lengthening in APD50 and APD80(P 0.05).10 μmol/L Propafenone resulted in a significant decrease in VDD and RPF(P 0.01),a notable decrease in MDP(P 0.05),APA(P 0.01) and Vmax(P 0.05),a marked lengthening in APD50(P 0.05) and APD80(P 0.01).10 μmol/L Adr led to a significant increase in VDD,RPF,Vmax,MDP and APA(P 0.05),a notable shortening in APD50(P 0.01).1 μmol/L Propafenone+10 μmol/L Adr resulted in a marked decrease in RPF,MDP,Vmax and APA(P 0.05),a notable decrease in VDD(P 0.01),a significant lengthening in APD50 and APD80(P 0.05).【Conclusion】 Propafenone could significantly decrease the autorhythmicity of left ventricular outflow tract and markedly alter the effects of Adr.
