摘  要:以黄鳝为材料,提取肝脏总RNA,经RT-PCR扩增出Hepcidin基因cDNA的开放阅读框(ORF)及3′端非编码区序列(GenBank序列号FJ436807),序列分析表明,黄鳝Hepcidin含有一个273 bp的ORF,编码90个氨基酸残基,有信号肽(24个残基)、前体肽(40个残基)和成熟肽(26个残基)。在前肽部分具有前肽转化酶典型的RX(K/R)R基元。同源性比对表明,黄鳝Hepcidin与其他鱼类的同源性在40%以上,是Hepcidin抗菌肽家族的新成员。Hepcidin's open reading frame(ORF) and 3' end UTR were amplified from the total RNA of liver of rice field eel by RT-PCR and RACE with specific primers based upon the homology DNA sequence FJ436807 in GenBank.The length of the cloned cDNA was 580 bp with a 273 bp ORF encoding 90 amino acids.The prepropeptide consisted of a signal peptide(24 amino acids),a prodomain(40 amino acids) and a mature peptide(26 amino acids).The propeptide contained the typical.Relement of propeptide invertase,mature peptide part contained 6 conserved cysteine residues that could form 3 intrachain disulfide bond.According to the alignment with other Hepcidins,the deduced amino acid sequence of rice field eel Hepcidin share high identity of 40% with those of other species,suggesting it was a new member of Hepcidin family.
