摘  要:目的探讨氯化镧对人大肠癌HT-29细胞增殖及肿瘤相关microRNA表达的影响。方法常规培养HT-29细胞,MTT法测定氯化镧对HT-29细胞的增殖影响,实量定量PCR法检测氯化镧作用HT-29细胞后microRNA let-7和miR34a的表达情况。结果 25μmol/L氯化镧可抑制HT-29细胞生长,肿瘤生长抑制率为:14.33%,且随着浓度增加(50、100μmol/L),肿瘤抑制率分别为:36.35%和64.52%,与对照组(0%)相比,差异显著,可见氯化镧可显著抑制大肠癌细胞HT-29的增殖(P<0.01)。实量定量PCR实验结果表明:低浓度氯化镧(25μmol/L、50μmol/L)对大肠癌HT-29细胞let-7表达的影响不明显(0.61±0.30,0.82±0.37),而100μmol/L氯化镧则可显著上调let-7的表达(2.64±0.57,与对照组相比P<0.05)。低浓度氯化镧(25μmol/L、50μmol/L)对大肠癌HT-29细胞miR34a表达的影响不明显(0.66±0.26,0.86±0.33),而100μmol/L氯化镧则可显著上调miR34a的表达(4.77±0.78,与对照组相比P<0.01)。结论一定浓度氯化镧可抑制人大肠癌细胞的增殖并上调抑癌相关microRNA的表达。Objective To investigate the effects of lanthanum chloride on the proliferation of human colorectal cancer HT-29 cells and the expression of tumor-associated microRNAs.Methods Human colorectal cancer HT-29 cells were cultured routinely in vitro.The inhibition effects of lanthanum chloride on cell-proliferation were determined by MTT method.The effects of lanthanum chloride on the expression of microRNA let-7 and miR34a in HT-29 cells were evaluated by real time quantitative PCR.Results Lanthanum chloride could inhibit the growth of HT-29 cells,and with the increacing of the dosage(25μmol/L,50μmol/L,100μmol/L),the tumor growth inhibition rate increased significantly(14.33%,36.35%,64.52%;P〈0.01).Meanwhile,the expression of let-7 and miR34a was not effected significantly by lower dosage of lanthanum chloride(25 and 50μmol/L).However,when HT-29 cells were treated by 100μmol/L lanthanum chloride,the expression of let-7 and miR34a was up-regulated significantly(2.64±0.57,4.77±0.78;P〈0.05).Conclusion The appropriate dosage of lanthanum chloride can inhibit HT-29 colorectal cancer cell growth in vitro,and can up-regulate the expression of tumor associated microRNA.
